摘 要:目的探讨羊栖菜褐藻糖胶(Sargassum fusiforme fucoidan,SFF)对表皮屏障损伤的修复作用。评价SFF对角质形成细胞增殖、迁移和分化的影响。方法使用BrdU和CCK8试剂盒分析SFF对HaCaT细胞的增殖作用影响;划痕法测定SFF对HaCaT细胞的迁移作用;流式细胞仪、qPCR和蛋白免疫印迹检测SFF对HaCaT细胞中mRNA表达和蛋白表达的影响。构建表皮屏障受损模型,通过经皮水散失、组织学观察等实验确定SFF对受损表皮屏障的体内修复作用。结果显示SFF以剂量依赖方式显著促进HaCaT细胞的增殖及迁移。此外,SFF能上调HaCaT细胞分化标志物的基因表达,通过激活ERK和JNK通路促进表皮角质形成细胞的增殖、迁移和分化。体内研究表明,与基质组相比,SFF处理组能促进表皮通透性屏障的恢复率,显著增加水合作用。结论SFF能够影响MAPK信号通路调节紊乱的角质形成细胞功能,作为治疗表皮屏障损伤的潜在药物或功能性化妆品添加成分。Objective To investigate the effect of fucoidan from Sargassum fusiforme fucoidan(SFF)on the repair of epidermal barrier injury,and to evaluate the effects of SFF on the proliferation,migration and differentiation of keratinocytes.Methods In the present study,the effects of SFF on epidermal permeability barrier recovery were examined.Besides,the effects of SFF on proliferation,migration and differentiation in keratinocytes were evaluated,which was critical to properly form the epidermal barrier.Moreover,the effects of SFF on epidermal permeability barrier recovery and normal skin were also evaluated.Results As revealed from our data,SFF could noticeably promote the proliferation of HaCaT cells under a dose-dependent effect.The treatment with SFF at a lower concentration(2.5μg·mL^(-1))also effectively promoted HaCaT cells to migrate.Moreover,SFF up-regulated the gene expression of differentiation markers in HaCaT cells,and promoted the proliferation,migration and differentiation of epidermal keratinocyte by activating ERK and JNK pathways.Furthermore,the recovery rate and hydration of epidermal permeability barrier after tape stripping were significantly increased in the SFF-treated group as compared with the vehicle-treated group(P<0.05).Conclusion SFF displays comprehensive functions to regulate the disordered keratinocytes and can be used as a potential ingredient in functional cosmetics and therapeutic agents to treat cutaneous disorders that are associated with abnormal epidermal barrier function.
