摘 要:目的探讨巴豆叶对脑缺血再灌注模型大鼠海马CA1区c-Jun-氨基末端激酶(JNK)通路的影响。方法将216只SD雄性大鼠按随机数表法分为假手术组、模型组、尼莫地平组、巴豆叶低、中、高浓度组,各组按取材时间点不同又分为1 d、3 d和7 d亚组,尼莫地平组灌胃尼莫地平混悬液1.08 g·L^(-1),巴豆叶低、中、高浓度组灌胃药物浓度分别为60 g·L^(-1)、120 g·L^(-1)、180 g·L^(-1),假手术组和模型组灌胃等体积蒸馏水,2次/d。以Garcia JH评分法进行神经功能评测,HE观察神经元形态;TUNEL法检测大鼠海马CA1区细胞凋亡;Western blot检测蛋白表达。结果1)神经功能评分:同一处理时间下,模型组和各干预组评分较假手术组均降低(P均<0.01);各干预组评分较模型组均升高,随着干预浓度的增加,评分也逐渐升高,尼莫地平组评分高于低、中浓度组(P均<0.01);除假手术组外,各组评分均随着干预时间的延长升高(P均<0.01)。2)HE染色:模型大鼠神经细胞出现不同程度的损伤;各药物干预组均能不同程度改善损伤;随着干预浓度的增加,修复程度也逐渐明显,尼莫地平组效果优于中、低浓度组;各干预组细胞形态均随着干预时间的延长明显改善。3)凋亡细胞计数:在各干预时间下,模型组及各干预组凋亡计数较假手术组均升高(P均<0.01);各干预组较模型组,计数均下降(P均<0.01);随着巴豆叶浓度的增加凋亡计数下降,且尼莫地平组高于中浓度组(P均<0.01)。4)Western blot检测:在各干预时间下,模型组及各干预组蛋白表达较假手术组均显著升高(P均<0.01);各干预组较模型组,蛋白表达下降(P均<0.01);随着浓度的增加蛋白表达下降,尼莫地平组低于中浓度组(P均<0.01);除假手术组外,各组大鼠蛋白表达均随着时间变化在1~7 d逐渐降低(P<0.01),且各时间点差异均有统计学意义(P均<0.01)。结论巴豆叶能下调JNK通路蛋白的表达,有效改善大鼠脑�Objective To investigate the effect of Croton leaf on c-Jun N-terminal kinase(JNK)pathway in hippocampal CA1 region of cerebral ischemia reperfusion rats.Methods A total of 216 male SD rats were divided into sham operation group,model group,nimodipine group,and soybean leaf with low,medium and high concentration groups according to random number table.Each group was further divided into 1 d,3 d and 7 d subgroups according to different sampling time points.Nimodipine group was given nimodipine suspension 1.08 g·L^(-1) by gavage.The intervention groups were orally administered with drug concentrations of 60 g·L^(-1),120 g·L^(-1),180g·L^(-1).Sham operation group and model group were given constant volume of distilled water twice a day.Neurological function was evaluated using the Garcia JH scoring method,and neuronal morphology was observed using HE;TUNEL assay was used to detect apoptosis in the CA1 region of the rat hippocampus;Western blot was used to detect protein expression.Results 1)Neurological scores:The scores of the model group and intervention groups were significantly lower than those of the sham operation group(P all<0.01);The scores of all intervention groups were higher than those of model group,and with the increase of intervention concentration,the scores of nimodipine group were higher than those of medium concentration group(P all<0.01);Except for the sham operation group,the scores of all groups were significantly increased with the extension of intervention time(P all<0.01).2)HE staining:nerve cells in model rats were damaged to different degrees;All the drug intervention groups could improve the injury to different degrees.With the increase of intervention concentration,the repair degree was gradually obvious,and the effect of nimodipine group was better than that of medium concentration group.The cell morphology of each intervention group significantly improved with the extension of intervention time.3)Apoptotic cell count:At each intervention time,the apoptotic cell count in model group
