摘 要:盐酸莫西沙星(moxifloxacin hydrochloride,MOX)是临床上广泛使用的喹诺酮类广谱抗菌药物,为探究纳米载体对其抗菌作用的影响,本研究利用表面吸附作用将MOX负载到纳米二氧化钛(titanium dioxide,TiO_(2))上,分析了纳米Ti O_(2)-MOX的化学表征及其对大肠杆菌(Escherichia coli)的抗菌活性和细胞病理学。纳米TiO_(2)与MOX以物理包埋的形式结合,随着处方中纳米TiO_(2)的含量从0.5 mg增加到1.0 mg,载药量增加了约20%,包封率增加了约40%,纳米TiO_(2)-MOX中的纳米TiO_(2)能帮助其有效成分MOX释放。合成的纳米TiO_(2)对大肠杆菌具有一定的抗菌效果(抗菌率为22.8%),经纳米TiO_(2)负载后的MOX对大肠杆菌的抗菌率由负载前的28.0%提高到73.4%。细胞病理学显微观察发现,MOX对大肠杆菌细胞膜的破坏不明显,纳米TiO_(2)和纳米TiO_(2)负载后的MOX可造成细胞膜明显破坏,进而造成菌体失活。本研究中合成的纳米TiO_(2)负载MOX后表现出了较好的载药和释放效果,且显著提高了MOX的抗菌活性,为今后纳米TiO_(2)作为药物载体用于临床更深层次的研究提供了科学依据。Moxifloxacin hydrochloride(MOX)is a broad-spectrum quinolone drug and is widely used in clinic as a antibacterial agent.In order to explore the effect of nanocarriers on its antibacterial effect,MOX was loaded onto nano titanium dioxide(TiO_(2))by surface adsorption in this study,and then the chemical characterization,antibacterial activity against Escherichia coli and pathological observation of the nano TiO_(2)-MOX were investigated.Nano TiO_(2) combined with MOX in the form of physical embedding.When the content of nano TiO_(2) increased from 0.5 mg to 1.0 mg,the drug loading and the encapsulation rate increased by about 20% and 40% respectively.Nano TiO_(2) in nano TiO_(2)-MOX could help release the effective components of MOX.The results of the in vitro activity against E.coli showed that the synthesized nano TiO_(2) had certain antibacterial activity(22.8%),and the antibacterial activity of MOX increased from 28.0% to 73.4% with the support of nano TiO_(2).Pathological observation also indicated that the damage of the cell membrane of E.coli was not obvious by MOX,but nano TiO_(2) and nano TiO_(2)-MOX could cause obvious damage to the cell membrane and then cause cell death.In this study,the synthesized nano TiO_(2)-MOX showed good drug loading and releasing effect,and significantly improved the antibacterial activity of MOX,which provides a scientific basis for the application of nano TiO_(2) as a new drug carrier in further clinical research.
