摘 要:目的分析新型冠状病毒(severe acute respiratory syndrome coronavirus 2,SARS-CoV-2)原型(Prototype)株、德尔塔(Delta)株及奥密克戎BA.1(Omicron BA.1)株在叙利亚金黄地鼠中初次感染和再感染的免疫反应和交叉保护效果。方法30只5~6周龄金黄地鼠随机分为初次感染组和再次感染组。初次感染组为腹腔感染SARS-CoV-2 Prototype、Delta或Omicron BA.1株,每种毒株感染5只地鼠。感染后第2、4、7、9、14天收集地鼠眼眶静脉丛血清样本,检测中和抗体水平;感染后第14天安乐死地鼠并收集动物肺、鼻甲组织检测病毒拷贝数。再次感染组在感染后第14天再次腹腔感染同类型毒株,并在再次感染后第14天安乐死地鼠并收集动物肺、鼻甲组织和血清样本。在整个实验过程中监测动物体质量。结果初次感染组中,所有感染组动物的体质量都增加,Omicron BA.1、Delta、Prototype感染组动物体质量分别由感染前的(101.29±9.13)、(109.04±2.08)、(113.51±3.19)g增加至感染后第14天的(142.67±7.34)、(150.48±9.36)、(144.96±7.34)g,分别增加了41.55%、38.04%和27.68%;初次感染组从第7天开始所有感染组均可检测到相应的中和抗体。除Omicron BA.1感染组在第14天中和抗体水平下降外,Prototype和Delta感染组抗体水平逐渐增加。在第14天时,Prototype、Delta和Omicron BA.1感染后诱导的动物体内中和抗体几何平均滴度(geometric mean titer,GMT)分别为499、844和48。Prototype、Delta和Omicron BA.1毒株再次感染组动物体质量增加(分别增加12.65%、12.60%和14.12%)都小于初次感染组,Prototype、Delta再次感染组地鼠的肺和鼻甲组织中的病毒拷贝数[M(Q_(25),Q_(75))]分别为3.17(2.77,3.83),3.03(2.58,3.51),0(0,3.68)和4.40(3.50,4.91);Omicron BA.1感染组病毒载量为阴性且显著降低(P<0.01)。Prototype和Delta之间有较强的交叉免疫原性,Prototype和Delta对Omicron BA.1变异株的交叉反应性较弱。结论Omicron新变异株具有较�Objective To analyze immunogenicity and cross-protective effects of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)Prototype,Delta and Omicron BA.1 strains in Syrian hamsters after primary infection and re-infection.Methods Thirty Syrian hamsters aged 5 to 6 weeks were randomly divided into primary infection group and re-infection group.The primary infection group was treated with abdominal infection with SARS-CoV-2 Prototype,Delta or Omicron BA.1,with each strain infecting 5 hamsters.Serum samples from the orbital venous plexus were collected 2,4,7,9 and 14 days after infection to detect neutralizing antibody levels.The hamsters in the primary infection group were killed on the 14th day after infection and their lung and nasal turbinate tissues were collected for measuring virus copy number.The hamsters in the re-infected group were re-infected with the same strain of SARS-CoV-2 in the abdominal cavity on the 14th day after the primary infection,and euthanized on the 14th day after re-infection to collect their lung,nasal turbinate tissue and serum samples.Animal body weight was monitored during the experiment.Results In the primary infection group,the body mass of all infected hamsters increased.The body mass of animals in the Omicron BA.1,Delta and Prototype infected sub-groups increased from(101.29±9.13),(109.04±2.08),and(113.51±3.19)g respectively before infection,to(142.67±7.34),(150.48±9.36),and(144.96±7.34)g respectively on the 14th day after infection,rising by 41.55%,38.04%,and 27.68%,respectively.In the primary infection group,corresponding neutralizing antibodies were detected in all the three sub-groups from day 7 onwards.On day 14,the neutralizing antibody level in Omicron BA.1 sub-group decreased(geometric mean titer,GMT:48),whereas the neutralizing antibody level in Prototype and Delta subgroups increased gradually(GMT:499 and 844,respectively).The increases of animal body mass in the re-infection group of Prototype,Delta and Omicron BA.1 strains(12.65%,12.60% and 14.12%,respecti
