摘  要:目的 探讨正常足月新生儿出生3个月内肠道菌群的定植规律。方法 收集20例正常足月新生儿及其孕母的临床资料,取新生儿出生后第1天(C0)、第3天(C1)、第1周(C2)、第1个月(C3)及第3个月(C4)的粪便标本,采用Illumina NovaSeq PE250进行16S核糖体RNA(16S rRNA)V3+V4区测序,采用生物信息学分析肠道菌群的定植规律。结果 正常足月新生儿粪便中肠道菌群的扩增子序列变异体(ASV)数目、Shannon指数于C0时最高;正常足月新生儿出生3个月内的肠道菌群以变形菌门(Proteobacteria)、厚壁菌门(Firmicutes)、放线菌门(Actinobacteriota)、拟杆菌门(Bacteroidota)为优势菌门,Proteobacteria相对丰度随日龄由高变低再逐渐增高,Firmicutes与之相反;正常足月新生儿生3个月内优势菌群在属水平的演变依次为罗尔斯通菌属(Ralstonia)、链球菌属(Streptococcus)、双歧杆菌属(Bifidobacterium),随日龄增加,Bifidobacterium相对丰度逐渐升高,C4时达44.18%且为优势菌属。结论 正常足月新生儿生3个月内肠道菌群的多样性、组成结构及相对丰度出现变化。Objective To investigate the colonization of intestinal flora in normal full-termneonates within 3 months after birth.Methods Clinical data of 20 normal full-term neonates and their pregnant mothers,and fecal specimens were collected from the 1 st day(C0),3 rd day(C1),1 st week(C2),1 st month(C3),and 3 rd month(C4)after birth.Illumina NovaSeq PE250 was used to sequencing and bioinformatic analysis of 16S ribosomal RNA(16S rRNA)V3+V4 region.Results The number of amplicon sequence variants(ASV)and Shannon index of intestinal flora in normal term neonates were the highest at C0.The postnatal intestinal flora of normal full-term neonates within 3 months after birth was dominated by Proteobacteria,Firmicutes,Actinobacteriota,and Bacteroidota.The relative abundance of Proteobacteria changed from high to low and then gradually increased with age,whereas Firmicutes were the opposite.Within 3 months after birth,the dominant bacteria in normal term neonates were Ralstonia,Streptococcus and Bifidobacterium at the genus level.With the increase of age,the relative abundance of Bifidobacterium increased gradually,reaching 44.18%at C4,and Bifidobacterium was the dominant bacterium.Conclusion The changes take place in diversity,composition and relative abundance of intestinal flora of normal full-term neonates within 3 months after birth.
