摘 要:目的构建自发性早产产妇胎盘与足月产妇胎盘差异表达基因cDNA文库, 初步分析自发性早产可能的分子机制。方法选取50例足月健康产妇胎盘和50例自发性早产产妇胎盘组织, 提取胎盘组织总RNA, 采用抑制性差减杂交(suppression subtractive hybridization, SSH)技术构建相应的cDNA差减文库, 进行克隆测序, 所得结果通过Blast软件与Genbank中的已知核苷酸序列进行查询和同源性比对, 分析可能和早产发生相关的分子机制。结果从正向及反向文库中随机挑选出200个克隆, 进一步筛选出80个差异表达基因片段。生物信息学方法分析显示, 两组差异表达基因功能涉及先天免疫、炎症、激素调节、营养代谢、应激及凋亡等方面。结论早产产妇胎盘组织中存在与先天免疫、炎症、激素调节、营养代谢、应激及凋亡等相关的基因表达变化, 其中GGT2基因很可能是1个新的导致自发性早产发生的基因, 其具体调控机制有待进一步深入研究。Objective To construct the cDNA library of differentially expressed genes in the placentas of spontaneous preterm and full-term,and to analyze the possible molecular mechanism of spontaneous preterm births.Methods Total RNA was extracted from placenta tissues of 50 full-term healthy women and 50 spontaneous preterm women,and corresponding cDNA subtractive libraries were constructed by suppressive subtractive hybridization(SSH)technology,and cloned and sequenced.The obtained results were queried and homologous compared with known nucleotide sequences in Genbank by Blast software.Results Bioinformatics analysis showed that the differentially expressed gene functions of the two groups involved innate immunity,inflammation,hormone regulation,nutrient metabolism,stress and apoptosis.Conclusions There are gene expression changes related to innate immunity,inflammation,hormone regulation,nutrient metabolism,stress and apoptosis in the placenta tissues of preterm mothers,suggesting that gamma-glutamyltransferase 2(GGT2)gene expression changes may promote the occurrence of spontaneous preterm birth.
