摘　　要：目的探究藏药青鹏骨伤灵复方乙醇提取物(QPEE)对大鼠关节炎的治疗作用及潜在机制。方法首先通过酊剂浸渍法获得QPEE,qRT-PCR检测其对IL-1β、TNF-α、IL-6和iNOS的mRNA表达的作用。免疫印迹法(WB)检测TLR4/p65-NFκB炎症通路相关蛋白的表达水平。通过构建胶原诱导关节炎(CIA)大鼠模型,评估QPEE对CIA大鼠的治疗作用。采用Micro-CT检测CIA大鼠股骨骨密度。结果在细胞水平上,QPEE对炎症因子IL-1β、TNF-α、IL-6和iNOS的mRNA表达具有显著抑制作用。在动物水平上,QPEE对CIA大鼠股骨骨密度具有显著改善的作用,并且对CIA大鼠脚踝肿胀具有显著的抑制作用。结论QPEE可能是通过抑制TLR4/p65-NFκB炎症通路活化,达到治疗CIA大鼠的作用。本研究为藏药青鹏骨伤灵复方在临床上治疗关节炎的应用提供了理论支持。OBJECTIVE To investigate the therapeutic effect and potential mechanism of Tibetan medicine QPEE on arthritis in rats.METHODS Firstly,QPEE was obtained by tincture immersion method,qRTPCR was used to detect the mRNA expression levels of IL1β,TNFα,IL6 and iNOS.The expression levels of TLR4/p65NFκB inflammatory pathway related proteins were detected by WB.Evaluate the therapeutic effect of QPEE on CIA rats by constructing a CIA rat model.MicroCT was used to detect femoral bone density in CIA rats.1 RESULTS At the cellular level,QPEE significantly inhibited the mRNA expression of inflammatory factor IL1β,TNFα,IL6 and iNOS.At the animal level,QPEE had a significant improvement effect on femoral bone density in CIA rats and a significant inhibitory effect on ankle swelling in CIA rats.CONCLUSION QPEE may have therapeutic effects on CIA rats by inhibiting the activation of the TLR4/p6NFκB inflammatory pathway.This study provides theoretical support for the clinical application of the Tibetan medicine Qingpeng Gushangling Compound in the treatment of arthritis.
