摘 要:目的:探讨电针对慢性压迫性损伤(CCI)模型大鼠神经病理性疼痛的缓解作用及相关机制。方法:选取雄性SD大鼠40只,随机数字表法分为假手术组、模型组、电针组与电针+激活剂组,每组10只。构建慢性压迫性损伤(CCI)大鼠模型,并按组别进行相应干预。测定大鼠痛觉敏感性,采用酶联免疫吸附测定(ELISA)法检测血清中IL-1β、IL-6和IL-8表达水平,RT-qPCR检测大鼠脊髓中CRMP2、Ubc9 mRNA及Western bolting检测大鼠脊髓中坍塌反应调节蛋白2(CRMP2)、泛素结合酶9(Ubc9)蛋白水平。结果:CCI大鼠模型建成后,各组大鼠PWLD均显著升高且>4 s,差异具有统计学意义(P<0.05);在给予大鼠相应干预后,与假手术组比较,模型组大鼠患健侧PWLD显著升高,差异具有统计学意义(P<0.05);与模型组比较,电针组和电针+激活剂组大鼠患健侧PWLD更低,差异具有统计学意义(P<0.05);与假手术组比较,模型组大鼠血清IL-1β、IL-6和IL-8水平升高,差异具有统计学意义(P<0.05);与模型组比较,电针组和电针+激活剂组大鼠血清IL-1β、IL-6和IL-8水平降低,差异具有统计学意义(P<0.05);与假手术组比较,模型组大鼠脊髓CRMP2、Ubc9 mRNA和蛋白表达水平更低,差异具有统计学意义(P<0.05);与模型组比较,电针组和电针+激活剂组大鼠脊髓CRMP2、Ubc9 mRNA和蛋白表达水平更高,差异具有统计学意义(P<0.05);电针+激活剂组各指标均优于电针组,差异具有统计学意义(P<0.05)。结论:电针可有效缓解CCI模型大鼠神经病理性疼痛,其作用机制可能是电针刺激穴位后引起机体CRMP2、Ubc9表达水平增加,抑制炎症反应,诱导镇痛。Objective:To explore the relieving effect and related mechanism of electro-acupuncture on neuropathic pain in chronic compression injury(CCI)model rats.Methods:40 male SD rats were randomly divided into the sham operation group,the model group,the electro-acupuncture group and the electro-acupuncture plus activator group,with 10 rats in each group.The CCI model was established in the rats and the corresponding intervention was carried out according to the groups.Pain sensitivity of rats was measured,and serum levels of IL-1β,IL-6 and IL-8 were detected by ELISA,the mRNA levels of CRMP2 and Ubc9 in rat spinal cord were detected by RT-qPCR,and the levels of collapse response regulatory protein 2(CRMP2)and ubiquitin-binding enzyme 9(Ubc9)protein in rat spinal cord were detected by Western blotting.Results:After the successful establishment of CCI rat model,PWLD of rats was significantly increased and was greater than 4's in each group(P<0.05).In terms of PWLD of the affected side and the unaffected side,it was higher in the model group than that in the sham operation group(P<0.05);it was lower in the electro-acupuncture group and the electro-acupuncture plus activator group than that in the model group(P<0.05).In terms of the serum levels of IL-1β,IL-6 and IL-8,they were significantly increased in the model group compared with those in the sham operation group(P<0.05);they were decreased in the electro-acupuncture group and the electro-acupuncture plus activator group compared with those in the model group(P<0.05).In terms of the mRNA and protein expressions of CRMP2 and Ubc9 in the spinal cord,they were lower in the model group than those in the sham operation group(P<0.05);they were higher in the electro-acupuncture group and the electro-acupuncture plus activator group than those in the model group(P<0.05).The improvements of the above indexes were more significant in the electro-acupuncture plus activator group than those in the electro-acupuncture group(P<0.05).Conclusion:Electro-acupuncture can effectively r
