摘  要:目的:观察电针预处理对永久性脑缺血大鼠不同时相、不同缺血区域钠钙交换体1(NCX1)表达的影响,并探讨其可能机制。方法:84只雄性SD大鼠随机分为假手术组、模型组和电针组,每组再分0、1、2和24 h亚组,每亚组各7只。电针刺激大鼠“百会”、双侧“内关”和“三阴交”,疏密波,频率2/15 Hz,强度1 mA,每次20 min,连续5 d。于末次电针后2 h采用线栓法制备大鼠永久性脑缺血模型,通过免疫组化染色及Western Blot观察电针预处理对不同时相(0、1、2和24 h)、不同缺血区域(缺血区、非缺血区)皮层NCX1阳性细胞数及蛋白表达的调控模式。结果:缺血区,3组间0 h时相NCX1蛋白表达差异无统计学意义(P>0.05);模型组NCX1蛋白表达于1、2和24 h均低于同时相假手术组,差异具有统计学意义(P<0.05);电针组NCX1蛋白表达于1 h、2 h显著高于同时相模型组差异具有统计学意义(P<0.05),于24 h降至最低,与同时相模型组,差异无统计学意义(P>0.05)。非缺血区,3组间0 h时相NCX1蛋白表达比较差异无统计学意义(P>0.05);与同时相假手术组比较,模型组NCX1蛋白表达于2 h、24 h显著降低,差异具有统计学意义(P<0.05),电针组可上调2 h、24 h时NCX1蛋白表达,与同时相模型组比较,差异具有统计学意义(P<0.05)。结论:脑缺血后缺血区、非缺血区皮层NCX1蛋白表达均呈现不同程度的降低,而电针预处理可以上调缺血区1 h、2 h NCX1蛋白及非缺血区2 h、24 h NCX1蛋白表达,可能是其脑保护作用的途径之一。Objective:To observe the effects of electro-acupuncture(EA)pretreatment on the expression of Na+/Ca 2+exchanger 1(NCX1)at different phases in ischemic regions of rats with permanent cerebral ischemia,and to explore its possible mechanism.Methods:84 male SD rats were randomly divided into the sham operation group,the model group and the EA group,and each group was subdivided into subgroups of 0 h,1 h,2 h and 24 h,with 7 rats in each one.EA stimulation was applied to Baihui(DU20),bilateral Neiguan(PC6)and Sanyinjiao(SP6),density wave,frequency 2/15 Hz,intensity 1mA,20 min each time for 5 consecutive days.Two hours after the last EA intervention,the rat model of permanent cerebral ischemia was prepared by suture-occluded method.Immunohistochemical staining and Western blot were used to observe the regulation mode of NCX1 positive cells and protein expression in the cortex of different phase(0 h,1 h,2 h,24 h)and different ischemic areas(ischemic and non-ischemic areas)by EA pretreatment.Results:In ischemic zone,there was no statistical difference in the expression of NCX1 protein at 0 h among the three groups(P>0.05);the expressions of NCX1 protein in the model group at 1 h,2 h and 24 h were lower than those of the same time point in the sham operation group(P<0.05);the expressions of NCX1 protein in the EA group were significantly higher than those in the model group at 1 h and 2 h(P<0.05),but the difference was not statistically significant compared with that in the model group at 24 h(P>0.05).In non-ischemic area,there was no statistical difference in the expression of NCX1 protein at 0 h among the three groups(P>0.05);NCX1 protein expressions were significantly decreased in the model group compared with those in the sham operation group at 2 h and 24 h(P<0.05);the expression of NCX1 proteins were up-regulated in the EA group than those in the model group at 2 h and 24 h(P<0.05).Conclusion:The expressions of NCX1 protein in ischemic and non-ischemic cortical areas are decreased in different degrees after cerebral
