摘  要:本研究利用无芒粳稻(Oryza sativa subsp.japonica Kato)品种‘日本晴’作为受体亲本,短芒籼稻(Oryza sativa subsp.indica Kato)品种‘9311’作为供体亲本,构建染色体片段代换系CSSL119,初步定位了一个新的芒发育相关基因AWN-3。AWN-3被定位在2号染色体分子标记M4和M5之间的1.2 Mb范围内;与‘日本晴’相比,CSSL119的平均芒长、芒着生率、二次枝梗数、穗粒数和单株产量均显著增加;RT-qPCR结果表明,与‘日本晴’相比,CSSL119中OsCKX2、An-1和An-2的表达量显著降低,而OsRR2、OsRR4、OsRR7、OsRR11和GAD1的表达量则显著增加,这表明AWN-3可能参与OsRR2、OsRR4、OsRR7、OsRR11和OsCKX2的表达而增加穗粒数,而芒发育的促进可能与GAD1表达的增加相关。本研究结果为AWN-3的基因克隆、功能分析和产量性状的遗传改良奠定了基础。In this study, using chromosome segment substitution line CSSL119 derived from an awnless japonica rice(Oryza sativa subsp. japonica Kato) cultivar ‘Nipponbare' and a short-awned indica rice(Oryza sativa subsp. indica Kato) cultivar ‘9311', AWN-3, a new gene related to awn development, was primarily mapped between the two markers M4 and M5 of chromosome 2, approximately 1.2 Mb apart. Compared with ‘Nipponbare', the average awn length, awn rate, number of secondary branches, grain number per spike and yield per plant of CSSL119 was significantly increased, respectively. The results of RT-qPCR showed that the expression of OsCKX2, An-1 and An-2 in CSSL119 was significantly lower than that in ‘Nipponbare', whereas the expression of OsRR2,OsRR4, OsRR7, OsRR11 and GAD1 was significantly increased. These indicated that AWN-3 might increase the number of grains per spike by promoting the expression of OsRR2, OsRR4, OsRR7 and OsRR11, and suppressing the expression of OsCKX2. Moreover, AWN-3 might affect the development of awn by promoting the expression of GAD1.These results of this study laid a foundation for AWN-3 gene cloning, functional analysis and genetic improvement of yield traits.
