摘 要:采用NaNO_(2)-Al(NO_(3))_(3)-NaOH法测定各组分总黄酮含量;DPPH、ABTS自由基清除及Fe^(3+)还原能力试验测定各组分抗氧化活性;Griees试剂法评价各组分抗炎活性。研究发现,变叶榕根提取物总黄酮含量为(161.26±2.74)mg/g,乙酸乙酯组分总黄酮含量最高,为(285.75±5.99)mg/g。乙酸乙酯组分对DPPH、ABTS自由基清除和Fe^(3+)还原能力的IC_(50)值分别为15.53、6.69和56.52μg/mL,对LPS诱导RAW264.7细胞释放NO的抑制活性的IC_(50)值为63.07μg/mL。结果表明,变叶榕根乙酸乙酯组分具有较明显的抗氧化抗炎活性,其化学成分及作用机制值得进一步研究。The contents of total flavonoids in variousfractions were determined by NaNO_(2)-Al(NO_(3))_(3)-NaOH assay.Antioxidant activity of the different fractions were evaluated by DPPH,ABTS free radical scavenging and reducing power measurement,and anti-inflammatory activity were evaluated by measuring NO that released from mice s abdominal macrophage RAW264.7 induced by lipopolysaccharide.The results revealed that the content of total flavonoids of ethanol extraction of Ficus variolosa roots was(161.26±2.74)μg/mL.The content of total flavonoids of ethyl acetate fraction was(285.75±5.99)μg/mL,which was significantly higher than that of any other fractions.EtOAc fraction had the most potent anti-oxidative and anti-inflammatory effect.IC 50 values for EtOAc fraction against DPPH,ABTS free radical,and reducing power were 15.53,6.69 and 56.52μg/mL,respectively.The IC_(50)values of the inhibitory activities of LPS-induced NO release in RAW264.7 cells was 63.07μg/mL.EtOAc fraction had obvious antioxidant and anti-inflammatory activities,and its chemical composition and mechanism of action deserved further study.
