摘 要:试验旨在开展奶牛群体高乳房炎抗性功能基因的筛选与验证,通过分析SNP位点多态性及其与体细胞数和产奶性状的相关性,探讨其对乳房炎的抗病情况。本研究对北京地区568头中国荷斯坦牛DCK、HIST1H2BK基因的多态性进行了检测,并对3个多态位点不同基因型与体细胞数、产奶性状进行了关联分析。结果表明,DCK基因的SNP位点6:g.86337334 A>G与体细胞数极显著相关(P<0.01),SNP位点6:g.86322040 C>T [rs43472176]与体细胞数也呈现极显著相关(P<0.01);HIST1H2BK基因的SNP位点23:g.31354269 T>G[rs41654340],与体细胞数呈显著关联(P<0.05),与乳脂率和乳蛋白率呈极显著关联(P<0.01)。本研究结果表明DCK、HIST1H2BK基因位点与体细胞数性状显著相关,其可能通过直接或间接的途径影响奶牛的乳脂率或乳蛋白率性状。本研究为荷斯坦牛后续的标记辅助选择奠定了良好的基础。The purpose of this experiment was to screen and verify the functional genes with high mastitis resistance in Chinese Holstein cows population.The gene frequencies of three mastitis related gene loci in Beijing were analyzed using sequenom technology.At the same time,the gene polymorphism and its correlation with Somatic Cell Count(SCC)and milk production traits were analyzed.In this study,the polymorphisms of DCK and HIST1H2BK genes in 568 Chinese Holstein cows in Beijing were detected,and the relationship of different genotypes of three SNPs with SCC and milk production traits was analized.The results showed that SNP 6:g.86337334 A>G of the DCK gene was highly significantly correlated with the number of SCC(P<0.01),and the SNP 6:g.86322040 C>T[rs43472176]was also highly significantly correlated with SCC(P<0.01).The SNP g.31354269 T>G[rs41654340]was significantly associated with SCC(P<0.05),and was extremely significantly associated with the rate of milk fat and milk protein(P<0.01).The results showed that DCK and HIST1H2BK genes could be used as candidate genes for marker assisted selection affecting mastitis traits in Chinese Holstein cows,and the above SNPs were significantly correlated with SCC traits.It may directly or indirectly affect the milk fat or protein content traits of cows.This study lays a solid foundation for subsequent marker assisted selection in Holstein cows.
