基于TGF-β1/Smads通路研究化纤中药颗粒干预纳米氧化镍所致肺纤维化的作用

摘  要:目的探讨化纤中药颗粒(HXF)对纳米氧化镍(NiO)所致的肺纤维化的干预效果和作用机制。方法将28只健康成年雄性SD大鼠随机分为对照组、纳米NiO组、纳米NiO+500 mg/kg HXF干预组和纳米NiO+1000 mg/kg HXF干预组。Mssson染色和羟脯氨酸(Hyp)含量检测确定大鼠肺组织胶原蛋白沉积情况,Western blot检测大鼠肺组织Ⅰ型胶原蛋白(Col-Ⅰ)、TGF-β1和Smad3蛋白的表达水平。结果与对照组相比,纳米NiO组大鼠肺组织大量胶原纤维沉积,Hyp含量和Col-Ⅰ蛋白表达升高,TGF-β1/Smads通路中TGF-β1和Smad3蛋白的表达均明显升高(F=10.568,F=4.593,F=28.354,F=31.533,F=17.236,P<0.05);与纳米NiO组相比,Masson染色结果显示HXF干预组大鼠肺组织胶原沉积减少,肺组织Hyp含量降低,Col-Ⅰ、TGF-β1和Smad3蛋白表达明显降低(P<0.05)。结论HXF可通过抑制TGF-β1/Smads信号通路干预纳米NiO所致的肺纤维化。Objective To study the intervention effect of Hua-Xian formula(HXF)on pulmonary fibrosis induced by nickel oxide nanoparticles(nano NiO),and to explore its possible mechanism.Methods Twenty-eight healthy adult SD male rats were randomly divided into control group,nano NiO group,nano NiO+500 mg/kg HXF group and nano NiO+1000 mg/kg HXF group.Mssson staining and hydroxyproline(Hyp)content detection were used to determine the collagen deposition in rat lungs,and the protein expression of type I collagen(Col-I),TGF-β1 and Smad3 were measured by Western blot.Results Compared with the control group,large amount of collagen fibers were deposited around the alveoli,as well,Hyp contents,Col-Ⅰ,TGF-β1 and Smad3 protein expression were increased in nano NiO group(F=10.568,F=4.593,F=28.354,F=31.533,F=17.236,P<0.05);Compared with the nano NiO group,the collagen fibers and Hyp contents were reduced by HXF,meanwhile,Col-Ⅰ,TGF-β1 and Smad3 protein levels were declined in HXF groups(P<0.05).Conclusion These findings indicated that HXF which could alleviate pulmonary fibrosis was involved in inhibiting TGF-β1/Smads pathway.
