摘  要:目的 探讨精索静脉曲张动物模型中精子特异性阳离子通道(CatSper)3、CatSper4的表达差异,以及精索静脉曲张导致的弱精子症与CatSper3、CatSper4的关系;并探讨CatSper抑制剂对附睾成熟精子的作用。方法 建立精索静脉曲张大鼠模型。剪碎附睾组织,离心收集附睾精子,对大鼠进行精子形态学分析。采用qRT-PCR和Western-blot检测大鼠精子中CatSper3、CatSper4的表达情况。加入10μmol/L的CatSper抑制剂NNC 55-0396与健康大鼠附睾成熟精子孵育24 h, ELISA法检测实验组和对照组精子中腺嘌呤核苷三磷酸(ATP)含量。结果 精索静脉曲张大鼠模型中畸形精子比例较对照组增高,精子数量明显减少。qRT-PCR检测结果可见,实验组的CatSper3、CatSper4 mRNA水平较对照组升高;但Western-blot检测发现,CatSper3、CatSper 4蛋白相对表达量较对照组明显降低。通过CatSper抑制剂NNC 55-0396处理附睾精子后,其ATP含量无显著变化。结论 精索静脉曲张可导致精子数量减少和畸形精子产生,降低精子中CatSper3、CatSper4蛋白的表达水平,但由于CatSper仅影响精子的活力,故认为CatSper3、CatSper4的蛋白表达下降不能解释弱精子症。Objective To explore the expression differences of cation channel of sperm(CatSper)3 and CatSper4 in animal models of varicocele as well as to determine the relationship between asthenospermia caused by varicocele and CatSper3 and CatSper4,and to investigate the effect of CatSper inhibitors on mature epididymal sperm.Methods A varicocele rat model was established under a microscope.The epididymal sperm was collected by cutting the epididymis tissue and centrifuged.The rats sperm morphology was analyzed.CatSper3 and CatSper4 in rat sperm were detected through qRT-PCR and Western-blot.The adenine nucleoside triphosphate(ATP)contents in sperm between the intervention and non-intervention groups were detected using ELISA after co-incubating mature sperm of epididymis in healthy rats with 10μmol/L NNC 55-0396 for 24 hours.Results The proportion of abnormal sperms increased,and the number of sperms decreased in the varicocele rat model.Increased mRNA of CatSper3 and CatSper4 was detected by qRT-PCR compared to the control group,whereas Western-blot analysis showed that the expression of CatSper3 and CatSper4 was significantly lower than that in the control group.The CatSper inhibitor NNC 55-0396 had no significant effect on ATP content in epididymal sperm.Conclusion Varicoceles can lead to a reduction in the number of sperms and increased abnormal sperms.Additionally,it can reduce the expressions of CatSper3 and CatSper4 proteins in the sperm.However,because CatSper only affects sperm motility,it is believed that the decreased protein expressions of CatSper3 and CatSper4 cannot be used to explain asthenospermia.
