摘 要:由果生刺盘孢Colletotrichumfructicola引起的苹果炭疽叶枯病是我国苹果上的一种新病害。前期通过组学分析发现一个在侵染早期表达的转录因子CfCrzA,本研究拟明确其在果生刺盘孢生长发育和致病中的作用。生物信息学分析显示,CfCrzA为刺盘孢属保守基因。采用同源重组方法获得2个敲除突变体。表型分析显示,突变体ΔCfCrzA与野生型WT生长速率无明显差别;致病性测定显示,ΔCfCrzA对苹果叶片和果实致病力显著下降。玻璃纸接种试验显示ΔCfCrzA孢子萌发率降低26%±3%,附着胞形成率降低43%±1%;叶片组织学观察发现ΔCfCrzA附着胞形成率降低60%±1%,但侵染泡囊和初生菌丝形成不受影响。培养发现ΔCfCrzA对细胞壁胁迫相关SDS的敏感性显著提升。这些结果表明转录因子CfCrzA为果生刺盘孢的关键致病因子,主要通过影响分生孢子萌发、附着胞形成等影响其致病力。Glomerella leaf spot caused by Colletotrichum fructicola is a new disease of apple in China.CfCrzA,a transcription factor expressed in the early stage of infection,has been previously found through omics analysis.The present study intends to clarify its role in the growth,development and pathogenicity of C.fructicola.Bioinformatics analysis showed that CfCrzA was a conserved gene in the Colletotrichum genus.Two deletion mutants of CfCrzA were obtained using homologous recombination.According to phenotypic analysis,ΔCfCrzA mutants had no significant difference in growth rate from wild type WT.The pathogenicity ofΔCfCrzA on apple leaves and fruits decreased significantly.Inoculation test on cellophane paper showed that the spore germination rate ofΔCfCrzA decreased by 26%±3%,and the appressorium formation rate reduced by 43%±1%.Leaf histological observation indicated that the appressorium formation rate ofΔCfCrzA decreased by 60%±1%,but the formation of infection vesicles and primary hyphae was not affected.Culture observation revealed that the sensitivity ofΔCfCrzA to cell wall stress-related SDS increased significantly.These results demonstrate that the transcription factor CfCrzA is a key pathogenic factor for C.fructicola,mainly influencing its pathogenicity through affecting conidial germination and appressorium formation.
