摘  要:药物靶点是一类在疾病进程中发挥关键作用的分子,药物靶点的发现和验证是新药研发的重要步骤,同时也深刻地影响后续药物研发的疗效、安全性等问题,通过设计能够抑制或促进靶点的药物以达到治疗疾病的效果,是目前药物开发的主要方式。在已知的药物靶点中,蛋白质类靶点是数量最多、研究最广泛的靶点。随着人们对于疾病的认识逐渐完善以及药物研发成本的不断加大,发现新的药物靶点对药物研发和临床应用有重要意义。近年来新的药物靶点发现以及验证方法持续开发、改进,大量潜在的靶点开始被研究人员关注。受益于计算机技术、组学技术及生物化学等实验方法的进步,越来越多的新兴药物蛋白靶点开始被研究并用于临床研究。这些技术在药物靶点发现的过程中发挥着重要的作用,同时也为药物作用机制、副作用发现等方面的研究提供帮助。文章综述药物蛋白靶点发现技术,为药物开发与应用提供理论依据。Drug target is a class of molecules that play a key role in the disease process,and the discovery and verification of drug targets is an important step in the research and development of new drugs,and also profoundly affects the efficacy and safety of subsequent drug research and development.Designing drugs that can inhibit or promote targets to achieve the effect of treating diseases is currently the main way of drug development.Among the known drug targets,protein-based targets are the most numerous and widely studied.With the gradual improvement of people's understanding of diseases and the increasing cost of drug research and development,the discovery of new drug targets is of great significance for drug development and clinical application.In recent years,new drug target discovery and verification methods have continued to be developed and improved,and a large number of potential targets have begun to be paid attention to by researchers.Benefiting from the advancement of computer technology,omics technology and biochemistry,more and more emerging drug protein targets have begun to be studied and used in clinical research.These technologies play an important role in the process of drug target discovery,and also provide assistance for the study of drug mechanism of action and side effect discovery.This article reviews the discovery technology of drug protein targets with the largest number of drug targets,which provides a theoretical basis for drug development and application.
