摘 要:【目的】筛选花椒鞘锈菌重寄生菌,并优化重寄生菌的最适产孢条件,为花椒锈病生物防治提供依据。【方法】利用形态特征及分子生物学对该菌株进行鉴定;采用单因素实验和Box-Behnken响应面法优化菌株HC30的产孢条件。【结果】从花椒叶锈病的夏孢子堆中,筛选获得菌株HC30对花椒鞘锈菌有重寄生作用,经形态学和分子生物学方法鉴定该菌为鞘柄锈柱隔孢菌。菌株HC30产孢条件优化结果显示,PSA培养基为最适培养基,蔗糖为最适碳源,硝酸钾为最适氮源,最佳培养条件为:温度25℃、氮源2.5%、碳源2.1%、pH 7,在此条件下添加花椒鞘锈菌夏孢子粉(0.5 g/L)的重寄生菌产孢量最好,为2.28×10^(8)spores/mL。【结论】从花椒叶锈病的夏孢子堆中筛选的菌株HC30对花椒鞘锈菌有强寄生作用,并得到了鞘柄锈柱隔孢菌的最佳产孢条件,为进一步研制生物菌剂提供理论依据。【:Objective】To screen the hyperparasite of Coleosporium zanthoxyli and optimize spore production conditions of the heavy parasitic bacteria,which provided a basis for the biological control of prickly ash rust.【Method】Using morphological characteristics and molecular biology to identify the strain.The single-factor and Box-Behnken response surface methods were used to optimize the spore-producing conditions of strain HC30.【Result】The strain HC30 was screened from the urediospores with rust and found to have hyperparasite effect on C.zanthoxyli,which was identified by morphological and molecular biological methods as Ramularia coleosporii.The optimization results of strain HC30 spore-producing conditions showed that PSA medium was the optimal medium,sucrose was the optimal carbon source,and potassium nitrate was the optimal nitrogen source.The optimal culture conditions were as follows:temperature of 25℃,nitrogen source of 2.5%,carbon source of 2.1%,pH of 7.Under these conditions,the best sporulation of 2.28×10^(8)spores/mL was achieved by the addition of C.zanthoxyli urediospores(0.5 g/L).【Conclusion】The strain HC30 screened from the urediospores with rust has strong hyperparasite effect on C.zanthoxyli,and the optimal spore production conditions of R.coleosporii are obtained,which provides a theoretical basis for further development of biological agents.
