摘 要:目的 探讨蒙药壮西-4调控miR-204对骨质疏松大鼠成骨细胞增殖的影响及机制。方法 将60只SD大鼠随机分为空白对照组、模型对照组、阳性对照组、低、中和高剂量组各10只。建模各组给予维A酸溶液灌胃建立骨质疏松模型,连续14 d。阳性对照组给予戊酸雌二醇灌胃,低、中、高剂量组分别给予不同剂量(50、100、200 mg/kg)蒙药壮西-4灌胃,空白对照组和模型对照组给予同体积的生理盐水灌胃,连续42 d。采用骨密度(BMD)仪测定大鼠第4腰椎及右股骨的BMD值,逆转录-聚合酶链反应(RT-PCR)血清miR-204表达量,酶联免疫吸附试验(ELISA)检测血清骨桥蛋白(OPG)、核因子κB受体活化因子配体(RANKL)含量。取MC3T3-E1细胞,分为对照组、促进组、抑制组和药物组,分别加入生理盐水、miR-204模拟物、miR-204抑制剂和蒙药壮西-4。采用RT-PCR法检测miR-204表达量,Western印迹检测RANKL、OPG蛋白表达量,噻唑蓝(MTT)检测细胞增殖情况,计算细胞存活率。结果 各组第4腰椎和右股骨BMD值、血清miR-204表达量、血清OPG含量差异有统计学意义(均P<0.05);空白对照组明显高于建模各组,模型对照组明显低于低、中、高剂量组、阳性对照组,阳性对照组和中剂量组明显高于低、高剂量组(均P<0.05)。各组血清RANKL含量差异有统计学意义(均P<0.05),空白对照组明显低于建模各组(均P<0.05),模型对照组明显高于低、中、高剂量组和阳性对照组(均P<0.05),阳性对照组和中剂量组明显低于低、高剂量组(均P<0.05)。各组miR-204表达量、OPG蛋白表达量、细胞存活率差异有统计学意义(均P<0.05),促进组和药物组明显高于对照组和抑制组,抑制组明显低于对照组(均P<0.05)。各组RANKL蛋白表达量差异具有统计学意义(均P<0.05),促进组和药物组明显低于对照组和抑制组,抑制组明显高于对照组(均P<0.05)。结论 蒙药壮西-4能增加骨质疏松大鼠BMD,其可能�Objective To investigate the effect and mechanism of miR-204 regulated by the Mongolian drug Zhuangxi-4 on the proliferation of osteoblasts in osteoporotic rats.Methods 60 SD rats were randomly divided into blank control group,model control group,positive control group,low,middle and high dose groups.Modeling groups were given retinoic acid solution by gavage for 14 consecutive days to establish the osteoporosis model.The positive control group was given estradiol valerate by gavage,and the low,medium and high dose groups were given different doses(50,100 and 200 mg/kg)of the Mongolian drug Zhuangxi-4 by gavage,while the blank control group and the model control group were given the same volume of saline by gavage for 42 consecutive days.The BMD values of the 4th lumbar vertebrae and right femur were measured by bone mineral density(BMD)instrument,the expression of serum miR-204 was determined by reverse transcription-polymerase chain reaction(RT-PCR),and the contents of serum osteoblastogenic polypeptide(OPG)and ribonucleic factorκB receptor-activated kinase ligand(RANKL)were detected by enzyme-linked immunosorbent assay(ELISA).MC3T3-E1 cells were divided into control,promoter,inhibitor and drug groups,and saline,miR-204 mimic,miR-204 inhibitor and Mongolian drug Zhuangxi-4 were added,respectively.The expression of miR-204 was detected by RT-PCR,the expressions of RANKL and OPG protein were detected by Western blotting,and the proliferation of cells was detected by thiazolyl blue(MTT),and the cell survival rate was calculated.Results The differences in BMD value,serum miR-204 expression and serum OPG content in the 4th lumbar vertebrae and right femur were statistically significant in all groups(all P<0.05);the above indicators in the blank control group were significantly higher than those in the modeling groups,the above indicators in model control group were significantly lower than those in low,medium,and high-dose groups and the positive control group,and above indicators in the positive control group and
