摘 要:目的运用数据挖掘联合网络药理学分析中医药治疗黄褐斑的用药规律及潜在作用机制。方法通过检索中国知网、万方及维普数据库关于中医药治疗黄褐斑的文献,建立处方数据库。运用SPSS20.0软件分析药物组方规律、关联规则和复杂网络分析,归纳中药在治疗黄褐斑用药规律及药物配伍。借助中药系统药理数据库和分析平台(TCMSP)数据库筛选出核心药对所含活性成分,DisGeNET和比较毒理基因组学数据库(CTD)获取黄褐斑疾病靶点,利用Cytoscape 3.9.1构建“化学成分-作用靶点”网络图,筛选出关键活性成分;利用STRING和Cytoscape获取核心靶点,使用DAVID数据库对核心靶点进行基因本体论(GO)与京都基因和基因组百科全书(KEGG)富集分析。结果整理出治疗黄褐斑方剂共221个,中成药166个,使用频率前五的药物即当归、地黄、柴胡、茯苓、白芍;关联规则数据显示当归-白芍-柴胡支持度与置信度最高。对“当归-白芍-柴胡”进行网络药理学分析,获取核心药对治疗黄褐斑的潜在作用靶点90个,GO富集和KEGG通路分析主要涵盖包括缺氧诱导因子(HIF)-1信号通路和磷脂酰肌醇-3-激酶(PI3K)-蛋白激酶B(Akt)信号通路等。结论通过数据挖掘和网络药理学方法初步获得了核心药对治疗黄褐斑多靶点、多途径的作用机制,为后续实验研究提供参考和依据。Objective Based on data mining and network pharmacology,to explore the compatibility and potential mechanism of traditional Chinese medicine in treating chloasma.Methods The literatures on the treat-ment of chloasma in the core journals from database was searched in CNKI,Wanfang Data and VIP databases to establish prescription database.The SPSS 20.0 software were used to analyzed the laws of drug composition and association rules,association traditional Chinese medicine in the treatment of chloasma the laws of drug use and drug compatibility.Core drug′s effective components were collected based on TCMSP.Relevant disease targets of chloasma were obtained by using the DisGeNet and CTD disease databases.The“active ingredient-potential target”network was constructed by Cytoscape 3.9.1 software for analysis,and the key active ingredients were screened out.Core targets were constructed by STRING and Cytoscape 3.9.1.GO and KEGG enrichment analysis of major targets were carried out through DAVID databases.Results A total of 221prescriptions and 166 traditional Chi-nese medicines were screened and summarized for the treatment of chloasma.The top five drugs used frequently were Angelica Sinensis,Rehmanniae Radix,Bupleurum,Poria and Paeoniae Radix Alba;Association rule data showed that the support and confidence of angelica sinensis-paeoniae Radix Alba-bupleurum were the highest.Network pharmacology analysis showed that“angelica sinensis-paeoniae Radix Alba-bupleurum”in the treatment of chloasma included 90 potential targets,GO enrichment and KEGG pathway analysis showed that the pathways included HIF-1 and PI3K-Akt signaling pathway.Conclusion Through data mining and network pharmacology methods,the action mechanism of core drugs on the treatment of chloasma is preliminarily obtained,which pro-vides reference and basis for subsequent experimental research.
