摘 要:目的探究白芷提取物介导转化生长因子-β(TGF-β)/Smad2通路对糖尿病大鼠感染性溃疡愈合效果的影响及相关机制。方法100只雄性SD大鼠采用随机数字表法分成对照组、模型组和白芷提取物低、中、高剂量组(3.8、7.5、15 mg/kg),每组20只。制备糖尿病大鼠感染性溃疡模型,观测各组创面情况(创面温度、红肿范围、溃疡面积和病理变化),对大鼠创面组织进行菌落计数,采用免疫组织化学法检测创面组织α平滑肌肌动蛋白(α-SMA)和血管内皮生长因子(VEGF)表达,蛋白质免疫印迹法检测创面组织中TGF-β1、Smad2和磷酸化Smad2(p-Smad2)蛋白表达。结果与模型组比较,白芷提取物中剂量组给药14、21 d(t_(14d)=4.377,t_(21d)=3.513,P<0.05)、高剂量组给药3、7、14、21 d(t_(3d)=3.049,t7d=5.166,t_(14d)=6.175,t_(21d)=5.270,P<0.05)创面温度降低,白芷提取物低、中剂量组给药7、14、21 d(低剂量:t7d=3.991,t_(14d)=8.860,t_(21d)=5.683,P<0.05;中剂量:t7d=7.052,t_(14d)=11.794,t_(21d)=9.848,P<0.05);高剂量组给药3、7、14、21 d(t_(3d)=3.237,t7d=11.356,t_(14d)=15.695,t_(21d)=15.187,P<0.05)创面愈合率升高(P均<0.05)。与模型组比较,白芷提取物低、中、高剂量组给药3、7、14及21d创面菌落数降低(低剂量:t_(3d)=2.580,t7d=2.614,t_(14d)=3.327,t_(21d)=4.923,P<0.05;中剂量:t_(3d)=4.519,t7d=3.505,t_(14d)=4.813,t_(21d)=6.141,P<0.05;高剂量:t_(3d)=5.201,t7d=5.147,t_(14d)=6.040,t_(21d)=9.632,P<0.05)。与模型组比较,白芷提取物中、高剂量组创面组织中α-SMA、VEGF表达升高(α-SMA:t=2.107、6.782,P<0.05;VEGF:t=2.653、6.002,P<0.05)。与模型组比较,白芷提取物中、高剂量组TGF-β1、p-Smad2/Smad2水平升高(TGF-β1:t=2.369、4.145,P<0.05;p-Smad2/Smad2:t=3.043、4.980,P<0.05)。结论白芷提取物可促进糖尿病大鼠感染性溃疡的愈合,有效清除创面细菌,可能与激活TGF-β/Smad2通路有关。Objective To explore the influences and related mechanism of Angelica dahurica extract on healing effect of infectious ulcers in rats with diabetes mellitus by mediating transforming growth factor-β(TGF-β)/Smad2 pathways.Methods According to random number table method,a total of 100 male SD rats were divided into control group,model group,low-dose,medium-dose and high-dose Angelica dahurica extract groups(3.8,7.5,15 mg/kg),20 cases in each group.The infectious ulcer models of diabetes mellitus rats were prepared.The wound situations(wound temperature,swelling range,ulcer area,pathological changes)were observed.The bacterial colonies of wound tissues were counted.The expressions ofα-smooth muscle actin(α-SMA)and vascular endothe-lial growth factor(VEGF)in wound tissues were detected by immunohistochemistry,and the expressions of TGF-β1,Smad2 and phosphorylated Smad2(p-Smad2)in wound tissues were detected by Western blot.Results Com-pared with model group,wound temperature was decreased in medium-dose Angelica dahurica extract group at 14 d and 21 d after administration(t_(14d)=4.377,t_(21d)=3.513,P<0.05)and high-dose group at 3 d,7 d,14 d and 21 d af-ter administration(t_(3d)=3.049,t7d=5.166,t_(14d)=6.175,t_(21d)=5.270,P<0.05),and wound healing rate was increased in low-dose and medium-dose groups at 7 d,14 d and 21 d after administration(low dose:t7d=3.991,t_(14d)=8.860,t_(21d)=5.683,P<0.05;medium dose:t7d=7.052,t_(14d)=11.794,t_(21d)=9.848,P<0.05)and high-dose group at 3 d,7 d,14 d and 21 d after administration(t_(3d)=3.237,t7d=11.356,t_(14d)=15.695,t_(21d)=15.187,P<0.05).Compared with model group,colony count was decreased in low-dose,medium-dose and high-dose Angelica dahurica extract groups at 3 d,7 d,14 d and 21 d after administration(low dose:t_(3d)=2.580,t7d=2.614,t_(14d)=3.327,t_(21d)=4.923,P<0.05;medium dose:t_(3d)=4.519,t7d=3.505,t_(14d)=4.813,t_(21d)=6.141,P<0.05;high dose:t_(3d)=5.201,t7d=5.147,t_(14d)=6.040,t_(21d)=9.632,P<0.05).Compared with model group,expressions ofα-SMA and VEGF were signif
