摘 要:目的:研究丹参-川芎药对干预血管性痴呆大鼠学习记忆能力的机制,及对海马神经元内的[Ca^(2+)]i、CaM、CaMPKⅡ活性的影响。方法:采用反复夹闭颈动脉、注射硝普钠降压、单侧永久性结扎颈动脉技术建立模型,采用水迷宫定位航行试验和空间探索试验测试大鼠在造模前、造模后、干预后的学习记忆能力,观察VD大鼠海马神经细胞[Ca^(2+)]i变化特征,检测海马组织中CaM、CaMPKⅡ的mRNA水平。结果:丹参-川芎高剂量组与尼莫地平组比较,差异无统计学意义(P>0.05)。丹参-川芎高剂量组穿越原平台区次数与尼莫地平组比较,差异无统计学意义(P>0.05),但优于丹参-川芎低剂量组(P<0.01)。模型组[Ca^(2+)]i荧光强度高于其他各组(均P<0.01),而尼莫地平组、丹参-川芎高剂量组的[Ca^(2+)]i荧光强度比较,差异无统计学意义(均P>0.05)。模型组的CaM mRNA、CaMPKⅡmRNA含量低于其他各组(均P<0.01),丹参-川芎高剂量组与尼莫地平组比较,差异无统计学意义(P>0.05)。结论:丹参-川芎药对能够阻止VD大鼠海马神经细胞[Ca^(2+)]i过度升高,且减少CaM、CaMPKⅡmRNA表达,能有效改善VD大鼠临床症状。Objective:To study effects of Danshen Chuanxiong couplet medicines on learning and memory abilities of vasculardementia rats and activities of[Ca^(2+)]i,CaM,and CaMPKⅡin hippocampal neurons.Methods:Model were established by repeatedly clamping carotid artery,injecting nitroprusside to reduce blood pressure,and unilateral permanent ligation of carotid artery.The learning and memory abilities of rats were tested before,after,and aftermodeling using a wateRmaze navigation test and spatial exploration test.The changes in[Ca^(2+)]i in hippocampal seminal cells of VD rats were observed,and mRNA levels of CaM and CaMPKⅡin hippocampal tissue were detected.Results:There was no statistical difference between the high-dose Danshen Chuanxiong group and the Nimodipine group(P>0.05).There was no statistically significant difference in the numberof times the high-dose Danshen Chuanxiong group crossed original platform area compared to the Nimodipine group(P>0.05).But it is superioRin the low-dose Danshen Chuanxiong group(P<0.01).In terms of[Ca^(2+)]i fluorescence intensity,the model group was significantly higheRthan otheRgroups(all P<0.01).There were no difference compared to the Nimodipine group,and the high-dose Danshen Chuanxiong group(P>0.05).The levels of CaM mRNA and CaMPKⅡmRNA in the model group were significantly lowerthan those in otheRgroups(all P<0.01).There was no difference between the high-dose Danshen Chuanxiong group and the Nimodipine group(P>0.05).Conclusion:Danshen Chuanxiong couplet medicines successfully prevented the excessive elevation of[Ca^(2+)]i in hippocampal neurons of VD rats and reduced expression of CaM and CaMPKⅡmRNA,while effectively improving clinical symptoms of VD rats.
