摘 要:为分析地方黄牛品种和杂交改良牛品种适应性特征的免疫机制,利用转录组学(RNA-seq)技术,以地方品种文山牛及杂交品种中国西门塔尔牛和云岭牛为研究对象,筛选种间差异表达的功能基因,挖掘免疫及抗病相关的重要调控基因。结果表明:文山牛脾脏组织的整体基因丰度显著高于中国西门塔尔牛和云岭牛,而2个杂交牛品种间无显著差异。基因表达量的相关性分析显示,相对于文山牛,中国西门塔尔牛与云岭牛的相关性较高。云岭牛与中国西门塔尔牛品种间差异基因数很少,仅为616个;而文山牛与中国西门塔尔牛以及文山牛与云岭牛品种间差异基因数明显增加,分别为4 503和3 520个。功能注释发现显著差异表达的基因多富集在免疫应答、对细菌的防御反应、对真菌的防御反应、对革兰阴性菌的防御反应、T细胞活化和中性粒细胞活化等通路上,其中ITK、IRF4、CD2、LAT等基因在地方牛中表达水平显著高于杂交牛,在免疫应答及疾病抵抗力过程中起着重要作用。To analyze the immunological mechanisms underlying the adaptive characteristics of local and hybrid improved cattle breeds, RNA-Seq technology was used to screen for differentially expressed functional genes between local breeds Wenshan cattle and crossbred Chinese Simmental and Yunling cattle, and to explore important regulatory genes related to immunity and disease resistance. The results showed that the overall gene abundance of Wenshan cattle was significantly higher than that of Chinese Simmental cattle. The expression levels of Chinese Simmental and Yunling cattle were close to each other with no significant difference. In addition, compared to Wenshan cattle, the gene expression had a higher correlation between Chinese Simmental cattle and Yunling cattle. Notably, the number of differentially expressed genes was remarkably lower in Yunling cattle vs Simmental cattle(616), compared to Wenshan cattle vs. Simmental cattle(4 503) and Wenshan cattle vs Yunling cattle(3 520). Functional annotation found that most of the significantly differentially expressed genes were related to immune responses, such as immune response, defense response to bacterium, defense response to fungus, defense response to Gram-negative bacterium, T cell activation and basophil activation pathways. Among them, genes such as ITK, IRF4, CD2, and LAT were significantly expressed in local cattle compared to hybrid cattle, which may play an important role in the cattle immune response and disease resistance.
