二至丸改善自然衰老大鼠卵母细胞线粒体功能的机制研究

摘  要:目的:研究二至丸对自然衰老大鼠卵母细胞线粒体改善的效应与机制。方法:选用SPF级SD雌性大鼠,随机分为8周龄青年对照组、20周龄老年对照组、20周龄二至丸用药低剂量组和高剂量组。用药30 d后,进行腹腔注射孕马血清促性腺激素(20 IU/只),2 d后给予人绒毛膜促性腺激素(10 IU/只),干预12 h后观察卵母细胞腺苷三磷酸(ATP)、活性氧(ROS)含量及线粒体的数目、形态,检测线粒体动态调控蛋白过氧化物酶体增殖物激活受体γ辅助活化因子1α(PGC-1α)、视神经萎缩蛋白1(OPA1)、PTEN诱导蛋白激酶1(PINK1)的蛋白表达水平。结果:和青年对照组相比,老年对照组卵巢卵母细胞ATP的含量减少(P<0.05),细胞内线粒体数量下降,结构紊乱,部分呈空泡化,PGC-1α、OPA1及PINK1蛋白水平下降(P<0.05);二至丸低、高剂量组可提高老年大鼠卵母细胞ATP的浓度(P<0.05),增加细胞内线粒体含量、改善结构及减少ROS的生成(P<0.05),促进PGC-1α、OPA1和PINK1的表达(P<0.05)。结论:二至丸可以改善衰老卵巢内的卵母细胞线粒体的结构和功能,提高线粒体自我更新以达到修复自然衰老卵母细胞的作用。Objective:To study the effect and mechanism of Erzhi Wan(Double Supreme Pill) on improving mitochondrial function in naturally aging rat oocytes.Methods:SPF-grade SD female rats were randomly divided into an 8-week-old youth control group,a 20-week-old elderly control group,two 20-week-old groups of low-dose of Erzhi Wan and high-dose of Erzhi Wan.After 30 days of medication,pregnant mare serum gonadotropin(20 IU/animal)was injected intraperitoneally.After 2 days,human chorionic gonadotropin(10 IU/animal)was administered.After 12 hours of intervention,adenosine triphosphate(ATP)in oocytes,reactive oxygen species(ROS)content,number and morphology of mitochondria were observed,mitochondrial dynamic regulatory protein peroxisome proliferator-activated receptor γ coactivator 1α(PGC-1α),optic atrophy protein 1(OPA1)and PTEN-induced protein kinase 1(PINK1)were measured.Results:Compared with the youth control group,the ATP content of ovarian oocytes in the elderly control group was reduced(P<0.05),and the number of intracellular mitochondria was significantly decreased,with structural disorder and partial vacuolization.PGC-1α,OPA1 and PINK1 proteins significantly decreased(P<0.05);both low-dose and high-dose of Erzhi Wan increased the concentration of ATP in oocytes of elderly rats(P<0.05),increased intracellular mitochondrial content,improve the structure,reduce the ROS generation(P<0.05),and promoted PGC-1α,OPA1 and PINK1 expressions(P<0.05).Conclusion:Erzhi Wan can improve the structure and function of mitochondria in aging oocytes,enhance the mitochondrial self-renewal,and achieve the effect of repairing naturally aging oocytes.
