摘 要:目的:根据真空润药机原理,对大黄进行真空置换软化,分析大黄真空置换软化过程中的质量变化,并探讨真空置换润药机替代传统大黄润制方法的可行性。方法:采用单因素和正交试验法考察大黄真空润药工艺,以2020版《中华人民共和国药典》“大黄”项下方法检测指标成分浸出物、总蒽醌及游离蒽醌,并与大黄传统润制方法后的质量进行比较。结果:真空置换润药机润制大黄的最佳工艺组合为浸泡时间6 h、润药温度65~70℃、润药时间60 min,采用真空润药软化后的大黄饮片含水量低、软化效果好、耗时短,且总蒽醌流失较少。结论:使用真空润药机润制大黄比传统软化方法润制大黄具有明显的优越性,保证了大黄饮片质量的均一性和稳定性,并提高了大黄成品含量和生产效率,真空置换润药替代传统浸泡润制大黄具有较好的可实施行性。Objective:According to the principle of Vacuum moistening machine,the Rhubarb is softened by gas phase replacement,optimizing the moistening process of Rhubarb,and The feasibility of replacing the traditional Rhubarb moistening method with vacuum replacement moistening machine.Methods:Single factor and orthogonal tests were used to investigate the moisturizing process,and the contents of extracts,total anthraquinones and free anthraquinones were measured by the determination method of Rhubarb effective components stipulated in the 2020 edition of the Chinese Pharmacopoeia for comprehensive evaluation,and the best optimization scheme was selected.And to verify and compare the traditional moistening method.Results:The best process combination for moistening Rhubarb by vacuum replacement moisturizing machine is soaking time 6h,moistening temperature 65~70℃,moistening time 60min;and the Rhubarb decoction pieces softened by vacuum moisturizing have low water content,good softening effect,Evenly softened.Conclusion:The use of vacuum moistening machine to moisten Rhubarb has obvious advantages over traditional softening method,which ensures the uniformity and stability of the quality of Rhubarb decoction pieces.
