摘 要:目的:基于数据挖掘和网络药理学方法探究中医药治疗原发性血小板减少性紫癜(ITP)的用药规律和潜在作用机制。方法:检索自2000年1月-2022年4月,CNKI、WanFang Data、VIP、CBM、PubMed、Cochrane Library及Web of Science数据库中中医药治疗ITP的相关文献,依据纳入排除条件收集其中药组方信息。以SPSS Modeler 18.0和SPSS Statistics 22.0作为统计分析工具进行用药规律分析。通过TCMSP、化学成分数据库等及SwissTargetPrediction平台获取核心药物组合的有效成分及靶点信息,通过GeneCards等疾病数据库获得ITP的相关疾病靶点。将药物靶点与疾病靶点取交集,运用Cytoscape 3.8.0软件构建潜在作用靶点-成分网络图。通过STRING 11.5和Cytoscape 3.8.0软件构建核心蛋白PPI网络。运用Metascape平台对交集靶点进行GO和KEGG富集分析。结果:最终获得184首中药处方,中药210味,使用频次最高的前六味中药分别为当归、仙鹤草、黄芪、生地黄、甘草、牡丹皮。关联规则分析显示,支持度最高的药对为生地黄、牡丹皮,系统聚类分析共聚出4个新处方。对当归、仙鹤草、黄芪、生地黄、牡丹皮进行网络药理学分析,获取药物靶点306个,疾病靶点1517个,交集靶点72个。最终获得关键成分为山柰酚、槲皮素、β-谷甾醇、白桦脂酸、木犀草素;筛选出核心靶点为AKT1、VEGFA、SRC、EGFR、CASP3等。GO分析得到生物过程1203条,包括蛋白质磷酸化的正调控、细胞表面受体蛋白酪氨酸激酶信号通路等;细胞组成78条,包括膜筏、膜微区等;分子功能95条包括蛋白酪氨酸激酶活性、蛋白激酶活性等,KEGG通路145条,与ITP相关性较大的通路为PI3K-Akt信号通路、癌症的MicroRNAs基因、MAPK信号通路等。结论:①中医药治疗ITP的药物,其用药类型以补益类最为常见,佐以清热凉血、收敛止血类药物等;②核心药物组合(当归、仙鹤草、黄芪、生地黄、牡丹皮)的主Objective:Based on data mining and network pharmacology methods,to explore the medication rules and potential mechanisms of traditional Chinese medicine in the treatment of idiopathic thrombocytopenic purpura(ITP).Methods:Searched by computer in databases of CNKI,WanFang Data,VIP,CBM,PubMed,Cochrane Library,and Web of Science since January 2000,about the relevant literature on the treatment of ITP by traditional Chinese medicine according to the inclusion and exclusion conditions.The medication rules were analyzed by SPSS Modeler 18.0 and SPSS Statistics 22.0 as statistical analysis tools.The effective components and target information of core drugs combinations are obtained through TCMSP,chemical composition database SwissTargetPrediction platform,etc.The disease targets of ITP are obtained through disease databases such as GeneCards and so on.Intersecting the drug targets and the disease targets,and constructing the potential target-effective components network by Cytoscape 3.8.0.Then build the PPI network through STRING 11.5 and Cytascape 3.8.0.Metascape platform is used to conduct GO and KEGG enrichment analysis on the intersection target.Results:184 prescriptions of traditional Chinese medicine and 210 prescriptions of traditional Chinese medicine were finally obtained.The top six herbs with the highest frequency were Angelica Sinensis Radix,Agrimonia Eupatoria,Hedysarum Multijugum Maxim,Radix Rehmanniae,licorice,and Cortex Moutan.Association rule analysis showed that the drug pairs with the highest support were Raw Rehmannia and Cortex moutan,and four new prescriptions were collected by systematic cluster analysis.Aanalysing the Angelica Sinensis Radix,Agrimonia Eupatoria,Hedysarum Multijugum Maxim,Radix Rehmanniae,and Cortex Moutan by network pharmacological method.306 drug targets,1517 disease targets,and 72 intersection targets were obtained in the end.The key components were obtained,including kaempferol,quercetin,β-Sitosterol,mairin,and luteolin.Core targets include AKT1,VEGFA,SRC,EGFR,CASP3,etc.By GO
