摘  要:目的:构建高脂血症防治模型,探讨炒白术-炒苍术对高脂血症大鼠水液代谢的影响。方法:48只雄性SD大鼠,随机分为空白对照组、模型对照组、依折麦布组(1 mg/kg)、炒白术-炒苍术组(6 g/kg)、炒白术组(3 g/kg)、炒苍术组(3 g/kg),各8只。空白对照组予以普通饲料喂养,其余组给予高脂饲料,连续6周。生化检测大鼠血脂胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C);ELISA检测血清胃泌素(GAS)、胃动素(MTL)、结肠Na^(+)-K^(+)-ATP酶;PCR及Western Blot检测结肠水通道蛋白(AQP)3、AQP9 m RNA及蛋白。结果:与空白对照组比较,模型对照组TC、LDL-C、MTL水平及AQP9 m RNA和蛋白水平显著升高(P<0.05,P<0.01),HDL-c、GAS、Na^(+)-K^(+)-ATP酶水平显著降低(P<0.01)。与模型对照组比较,炒白术-炒苍术组TC、TG、LDL-C水平及AQP9 m RNA和蛋白水平显著降低(P<0.05,P<0.01),GAS、Na^(+)-K^(+)-ATP水平显著升高(P<0.05,P<0.01)。结论:炒白术-炒苍术可能是通过调节Na^(+)-K^(+)-ATP酶和AQPs介导的水液代谢来防治高脂血症的。Objective:To construct a model for the prevention and treatment of hyperlipidemia,and to investigate the effects of fried atractylodes-fried atractylodis on aqueous metabolism in rats with hyperlipidemia.Methods:A total of 48 male SD rats were divided into blank control group,model control group,Ezetimibe group(1 mg/kg),fried atractylodes-fried atractylodis group(6 g/kg),fried atractylodes group(3 g/kg)and fried atractylodis group(3 g/kg),with 8 rats in each group.The blank control group was fed with ordinary diet,and the rest of the group was given high-fat diet for 6 consecutive weeks.Biochemical assays of blood lipid cholesterol(TC),triglyceride(TG),high-density lipoprotein cholesterol(HDL-C)and low-density lipoprotein cholesterol(LDL-C)in rats.ELISA was used to detect serum gastrin(GAS),motilin(MTL)and colonic Na^(+)-K^(+)-ATPase.PCR and Western Blot were used to detect colonic aquaporin(AQP)3 and AQP9 mRNA and protein.Results:Compared with the blank control group,the levels of TC,LDL-C,MTL,and AQP9 mRNA and protein in the model control group were significantly increased(P<0.05,P<0.01).The levels of HDL-C,GAS,Na^(+)-K^(+)-ATP were significantly decreased(P<0.01).Compared with the model control group,the levels of TC,TG,LDL-C,and the mRNA and protein levels of AQP9 in the fried atractylodes-fried atractylodis group were significantly decreased(P<0.05,P<0.01),the levels of GAS and Na^(+)-K^(+)-ATP were significantly increased(P<0.05,P<0.01).Conclusion:Fried atractylodes-fried atractylodis may be used to prevent hyperlipidemia by regulating Na^(+)-K^(+)-ATPase and AQPs-mediated water-liquid metabolism.
