摘  要:目的:以阿尔茨海默病(AD)模型线虫为模式生物,探究霜后连翘叶醇提物(AFLF)的抗β-淀粉样蛋白(Aβ)毒性作用,并探讨其机制。方法:通过检测AFLF对AD模型线虫寿命、瘫痪时间、趋化能力、Aβ沉积、活性氧水平和氧化应激相关基因(daf-16、skn-1、hsf-1、ctl-1、sod-3)表达水平的影响,来阐明AFLF抗Aβ毒性作用及机制。结果:与对照组比较,5mg/mLAFLF能够显著延长AD模型线虫的寿命(P<0.01),减少Aβ诱导的瘫痪毒性和趋化性缺陷,减少Aβ沉积,并显著提高daf-16、sod-3、hsf-1、skn-1和ctl-1基因的表达水平(P<0.01)。结论:AFLF可通过减少氧化应激反应,抑制Aβ的沉积,从而减轻AD模型线虫Aβ诱导的毒性。Objective:To explore the anti-amyloidβ(Aβ)toxicity of alcohol extract of Forsythia suspense leaves after frost(AFLF)in Alzheimer’s disease(AD)model nematode,and to discuss its mechanism.Methods:The effects of AFLF on lifespan,paralysis time,chemotaxis,Aβdeposition,reactive oxygen species and expression levels of oxidative stress-related genes(daf-16,skn-1,hsf-1,ctl-1,sod-3)in AD model nematodes were examined to elucidate the anti-Aβtoxicity of AFLF and its mechanism.Results:Compared with the control group,5 mg/mL AFLF could significantly prolong the life span of AD model nematode(P<0.01),reduce Aβ-induced paralytic toxicity and chemotactic defects,reduce Aβdeposition,and significantly increase the expression of daf-16,sod-3,hsf-1,skn-1 and ctl-1(P<0.01).Conclusion:AFLF can reduce Aβ-induced toxicity in AD model nematodes by reducing oxidative stress response and inhibiting Aβ-deposition.
