摘  要:通过对我国地方猪种与国外猪种比较,筛选与生长发育性状相关的差异候选基因,探索猪种之间生长发育差异的分子机制。分别选择5头内江猪和大约克夏猪进行全基因组重测序,以10 kb滑动窗口和1 kb步长计算群体间遗传分化指数(Fst)平均值,并依据Fst筛选差异SNPs。最终在内江猪与大约克夏猪的比较中分别鉴定出48924个非同义SNP,它们高度影响2490个基因。同时,在乙酰辅酶A乙酰转移酶1(ACAT1)、胰岛素样生长因子受体2(IGF2R)和胰岛素样生长因子2 mRNA结合蛋白3(IGF2BP3)基因中检测到3个非同义SNP,这可能影响乙酰辅酶A向乙酰乙酰辅酶A的转化和胰岛素信号通路的正常功能。本研究结果将为探索决定猪表型特征的遗传差异提供基础信息。The study aimed to explore the molecular mechanisms underlying the growth and developmental differences among these pig breeds.Five Neijiang(NJ)pigs and approximately five Yorkshire(YS)pigs were selected for whole-genome resequencing.Differential single-nucleotide polymorphisms(SNPs)were identified through a 10 kb window sliding in 1 kb increments using the Fst method.Ultimately,a total of 48924 non-synonymous single-nucleotide polymorphism sites(nsSNPs)were discovered between NJ and YS pjgs,significantly impacting 2490 genes.Moreover,three nsSNPs were identified in the genes ACAT1,IGF2R and IGF2BP3,potentially affecting the normal functioning of insulin signaling pathways and the conversion of acetyl-CoA to acetoacetyl-CoA.In conclusion,these findings contributed essential knowledge about genetic variations influencing phenotypic features in pigs.
