摘 要:目的建立测定炒榧子仁饮片中油酸和亚油酸含量的高效液相色谱法。方法色谱柱为Thermo Hypersil Gold C18柱(250 mm×4.6 mm,5μm),流动相为乙腈-0.1%磷酸水溶液(78∶22,V/V),流速为1.0 mL/min,柱温为30℃,检测波长为203 nm,进样量为10μL。结果亚油酸和油酸的进样量分别在0.2823~2.2582μg、0.1159~0.9274μg范围内与峰面积积分值线性关系良好(R2=1.0000,1.0000,n=8);精密度、稳定性、重复性试验结果的RSD均小于2.0%;加样回收率分别为99.83%和97.95%,RSD分别为1.79%和1.67%(n=9)。除含量明显偏低和明显偏高的各1批样品外,其余11批样品中亚油酸和油酸的总含量为(0.65±0.34)%。结论该方法操作简便、结果准确,可用于炒榧子仁饮片中油酸和亚油酸的含量测定。建议炒榧子仁饮片中油酸和亚油酸的总含量不得低于0.30%。Objective To establish a high-performance liquid chromatography(HPLC)method for the content determination of oleic acid and linoleic acid in Torreyae Semen Tostum.Methods The chromatographic column was Thermo Hypersil Gold C18 column(250 mm×4.6 mm,5μm),the mobile phase was acetonitrile-0.1%phosphoric acid aqueous solution(78∶22,V/V),the flow rate was 1.0 mL/min,the column temperature was 30℃,the detection wavelength was 203 nm,and the injection volume was 10μL.Results The linear ranges of linoleic acid and oleic acid were 0.2823-2.2582μg and 0.1159-0.9274μg(R2=1.0000,1.0000,n=8),respectively.The RSDs of precision,stability,and repeatability test results were all lower than 2.0%.The average recoveries of linoleic acid and oleic acid were 99.83%and 97.95%,with RSDs of 1.79%and 1.67%(n=9),respectively.Except for one batch of samples with significantly lower content and one batch with significantly higher content,the total content of linoleic acid and oleic acid in the remaining 11 batches of samples was(0.65±0.34)%.Conclusion This method is simple and accurate,which can be used for the content determination of oleic acid and linoleic acid in Torreyae Semen Tostum.It is recommended that the total content of oleic acid and linoleic acid in Torreyae Semen Tostum should not be lower than 0.30%.
