摘 要:目的观察三七总皂苷(PNS)对过氧化氢(H_(2)O_(2))诱导人肾小管上皮细胞(HK-2)的氧化应激损害的保护作用及其机制。方法培养HK-2并分组:正常组(不加任何干预因素),H_(2)O_(2)组(在细胞培养体系中加入终浓度为400μmol/L的H_(2)O_(2),培养6 h)和PNS组(分为3组:先在细胞培养体系中分别加入PNS 25μg/ml、50μg/ml和100μg/ml,培养4 h,再加入终浓度为400μmol/L的H_(2)O_(2),继续培养6 h),观察H_(2)O_(2)刺激HK-2发生凋亡情况。结果与正常组相比,H_(2)O_(2)组细胞存活率降低为(45.67±2.52)%、细胞凋亡率升高至(19.23±1.63)%(均P<0.01),而PNS组各组的细胞存活率分别为(55.12±2.33)%、(65.37±4.72)%和(83.46±1.52)%,细胞凋亡率分别降低至(15.53±0.70)%、(12.53±1.30)%和(7.17±0.35)%,50μg/ml和100μg/ml PNS组的HK-2细胞内活性氧(ROS)水平分别为(845.7±17.79)和(353.3±26.1);3个PNS组中,随着PNS剂量增加,超氧化物歧化物(SOD)和谷胱甘肽过氧化物酶(GSH-Px)的活性增加,丙二醛(MDA)的生成减少,与H_(2)O_(2)组相比,差异均存在显著统计学意义(均P<0.01)。结论PNS能通过增强细胞内抗氧化酶活力,保护HK-2细胞对抗氧化损伤,随着剂量增加,作用效果增强。Objective To observe the protective effect of Panax notoginseng total saponins(PNS)on oxidative stress damage induced by hydrogen peroxide(H_(2)O_(2))in human renal tubular epithelial cells(HK-2)and its mechanism.MethodsHK-2 was cultured and divided into experimental groups:Normal group(without any intervention factors),H_(2)O_(2) group(400μmol/L H_(2)O_(2) was added into the cell culture system for 6 h)and PNS group(which was divided into three groups:PNS 25μg/ml,50μg/ml and 100μg/ml were added into the cell culture system for 4 h,and then 400μmol/l H_(2)O_(2) was added into the cell culture system for 6 h),and the apoptosis of HK-2 induced by H_(2)O_(2) was observed.ResultsCompared with the normal group,the cell survival rate of H_(2)O_(2) group decreased to(45.67±2.52)%and the cell apoptosis rate increased to(19.23±1.63)%(all P<0.01),while the cell survival rate of PNS group was(55.12±2.33)%,(65.37±4.72)%and(83.46±1.52)%,and the apoptosis rate was reduced to(15.53±0.70)%,(12.53±1.30)%and(7.17±0.35)%,respectively.The levels of reactive oxygen species(ROS)in HK-2 cells in 50μg/ml and 100μg/ml PNS groups were(845.7±17.79)and(353.3±26.1),respectively.With the increase of PNS dose,super oxide dismutase(SOD)and glutathione peroxidase(GSH-Px)activities increased and malonic dialdehyde(MDA)production decreased in the 3 PNS groups,compared with the H_(2)O_(2) group,the differences were statistically significant(all P<0.01).ConclusionsPNS can protect HK-2 cells against oxidative damage by enhancing the activity of intracellular antioxidant enzymes,the effect is enhanced with the increase of the dose.
