摘  要:为了解我国部分地区猪场猪轮状病毒(porcine rotavirus, PoRV)的流行情况及分子特征,对2021-2022年我国部分省份的86个规模化猪场的6 472份腹泻样品进行检测,用RT-PCR方法调查PoRV的流行率,并对PoRV阳性样本进行VP7和VP4基因扩增、测序,使用MegAlign软件进行同源性分析,利用MEGA11.0构建系统进化树。结果显示,PoRV的样品阳性率为27.89%(1 805/6 472),猪场阳性率为65.12%(56/86);从98份阳性PoRV样本中扩增出76个VP7片段,G9为优势基因型(42.11%),基因型G5、G26、G3、G4、G1、G2和G11各占26.32%、9.21%、6.58%、6.58%、3.95%、2.63%和2.63%。扩增出的35个VP4基因片段,以P[13]型为主,占57.14%;其次为P[7]、P[23]、P[3]和P[6]型,分别占20%、14.29%、5.71%和2.86%。35个毒株成功鉴定出G/P基因型,G9P[13](28.57%)为优势组合基因型,其他基因型为G5P[13](11.43%)、G4P[13](8.57%)、G9P[23](8.57%)、 G26P[13](5.71%)、G1P[7](5.71%)、G26P[3](5.71%)、G5P[7](5.71%)、G3P[7](2.86%)、G11P[23](2.86%)、G5P[23](2.86%)、G11P[7](2.86%)、G3P[13](2.86%)、G5P[6](2.86%)和G4P[7](2.86%),其中G11P[7]为国内首次鉴定到的基因型。综上,目前PoRV的流行率高且基因型复杂,优势组合基因型为G9P[13]。该结果丰富了PoRV的分子流行病学资料,对我国猪PoRV感染的防控具有重要意义,也为新型疫苗研发提供了参考依据。In order to understand the prevalence and molecular characteristics of porcine rotavirus(PoRV)in some pig farms in China,in the period from 2021 to 2022,6472 diarrhea samples were collected from 86 large-scale pig farms in some provinces of China and were studied.The prevalence of PoRV was investigated using RT-PCR,and the VP7 and VP4 genes in the PoRV positive samples were amplified and sequenced.Then,homology analysis was performed using the MegAlign software,and a phylogenetic tree was constructed using MEGA110 A total of 1805 out of 6472 diarrhea samples were positive for PoRV,with a positive rate of 2789%(1805/6472)of all the samples and 6512%(56/86)in the pig farms.76 VP7 fragments were amplified from 98 positive PoRV samples,G9(4211%)was the dominant genotype,followed by G5,G26,G3,G4,G1,G2,and G11 which accounted for 2632%,921%,658%,658%,395%,263%,and 263%of the samples,respectively.The P[13]genotype(5714%)was dominant among the 35 P types,followed by P[7],P[23],P[3],and P[6]types,accounting for 20%,1429%,571%,and 286%,respectively.Furthermore,the G/P com-bination genotype was successfully identified in the 35 strains,and the dominant combination genotype was G9P[13](2857%),the other weaker combinations were G5P[13](1143%),G4P[13](857%),G9P[23](857%),G26P[13](571%),G1P[7](571%),G26P[3](571%),G5P[7](571%),G3P[7](286%),G11P[23](286%),G5P[23](286%),G11P[7](286%),G3P[13](286%),G5P[6](286%),and G4P[7](286%).Obviously,G11P[7]was first identified in China.Taken together,the current prevalence of PoRV was high and the genotypes were complex,with the dominant combina-tion genotype being G9P[13].The results of this study enriched the molecular epidemiology data of PoRV,which would be of great signifi-cance for the prevention and control of PoRV infection in pigs in China,and would also provide reference for the development of new vac-cines.
