摘 要:目的探究黄芪甲苷调节白介素-10(IL-10)/β-内啡肽(β-EP)信号通路对腰椎间盘突出症(LDH)大鼠神经根损伤的影响。方法随机选择10只大鼠记为假手术组(Sham组),其余大鼠通过移植髓核构建LDH大鼠模型,将造模成功大鼠随机分为LDH组、黄芪甲苷组(20 mg/kg黄芪甲苷)、AS10组(0.5 mg/kg IL-10/β-EP信号通路抑制剂AS10)、黄芪甲苷+AS10组(20 mg/kg黄芪甲苷+0.5 mg/kg AS10),每组10只,Sham组和LDH组给予等量0.9%氯化钠溶液。机械刺激敏感性实验以及热刺激敏感性实验检测大鼠机械缩足阈值(PWT)以及热痛阈(TWL);ELISA法检测血清白介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)水平;免疫荧光染色测定小胶质细胞和星形胶质细胞活性;TUNEL染色检测细胞凋亡;Western blot检测IL-10、β-EP蛋白表达。结果与Sham组比较,LDH组PWT值、TWL值、IL-10、β-EP蛋白水平显著减小(P<0.05),细胞凋亡率、TNF-α、IL-1β水平、离子钙接头蛋白分子1(Iba1)、胶质纤维酸性蛋白(GFAP)阳性细胞数量均显著升高(P<0.05);与LDH组比较,黄芪甲苷组PWT值、TWL值、IL-10、β-EP蛋白水平显著升高(P<0.05),细胞凋亡率、TNF-α、IL-1β水平、Iba1、GFAP阳性细胞数量均显著降低(P<0.05),而AS10组趋势相反(P<0.05);AS10组逆转了黄芪甲苷对LDH大鼠神经根损伤的改善作用。结论黄芪甲苷可能通过激活IL-10/β-EP信号通路减轻大鼠炎性反应,进而减轻LDH大鼠神经根损伤。Objective To investigate the impact of astragaloside IV on nerve root injury in rats with lumbar disc herniation(LDH)by regulating the interleukin-10(IL-10)/β-endorphin(β-EP)signaling pathway.Methods Ten rats were randomly selected and included in the sham group.The remaining rats were transplanted with the nucleus pulposus to construct a LDH rat model,followed by the grouping into LDH group,astragaloside IV group(20mg/kg astragaloside IV),AS10 group(0.5mg/kg IL-10/β-EP signaling pathway inhibitor AS10),and astragaloside IV+AS10 group(20mg/kg astragaloside IV+0.5mg/kg AS10),with 10 rats in each group.Rats in sham group and LDH group were given an equal amount of 0.9%NaCl solution.Mechanical sensitivity and thermal sensitivity tests were applied to detect the mechanical withdrawal threshold(PWT)and thermal withdrawal threshold(TWL),respectively.The enzyme-linked immunosorbent assay(ELISA)was applied to detect serum levels of interleukin-1β(IL-1β)and tumor necrosis factor-α(TNF-α).Immunofluorescence staining was applied to determine the activities of microglia and astrocyte.The terminal deoxynucleotidyl transferase biotin-dUTP nick end labeling(TUNEL)staining was applied to detect cell apoptosis.Western blot was applied to detect the protein expressions of IL-10 andβ-EP.Results Compared with those of the sham group,rats in LDH group presented significantly lower PWT,TWL,and protein levels of IL-10,andβ-EP in the LDH group(P<0.05),but significantly higher apoptosis rate,serum levels of TNF-αand IL-1β,and the numbers of ion calcium junction protein molecule 1(Iba1)and glial fibrillary acidic protein(GFAP)-positive cells(P<0.05).Compared with those of the LDH group,rats in the astragaloside IV group presented significantly higher PWT,TWL,and protein levels of IL-10 andβ-EP(P<0.05),but significantly lower apoptosis rate,serum levels of TNF-αand IL-1β,and the numbers of Iba1 and GFAP-positive cells(P<0.05),while the trends in the AS10 group were opposite(P<0.05).AS10 group reversed the improvement effect o
