摘 要:小麦叶锈病是由隐匿柄锈菌(Puccinia triticina,Pt)侵染的一种气传性真菌病害,对小麦造成严重的产量损失。Pt产生的吸器是产生效应因子的主要场所,而效应因子是Pt致病的关键。本研究基于早期构建的小麦叶锈菌单胞菌系13-5-28-1(JHKT)在休眠孢子、萌发夏孢子和侵染6 d的转录组文库,在系统筛选候选效应因子的基础上,选取两个候选效应因子Pt12448和Pt34354进行克隆,利用qRT-PCR分析候选效应因子的表达模式,结果发现其中Pt12448基因表现为后期诱导上调,Pt34354基因则为前期诱导上调。研究结果将推进对小麦叶锈菌致病机制的研究进程。Wheat leaf rust,caused by Puccinia triticina(Pt)is an airborne fungal disease in wheat worldwide,which can cause significant yield losses to the wheat production in the world.The haustoria of Pt is the main place where the effector proteins were secreted,and the effector protein is the key of the pathogenetic of Pt.The transcriptome librarie of wheat leaf rust races 13-5-28-1(JHKT)was constructed in the stages of dormant spores,spore germination and 6 d infection period.Based on the systematic screening of candidate effectors,two candidate effectors,Pt12448 and Pt34354,were selected for cloning,and the expression patterns of the candidate effectors were analyzed by qRT-PCR.The results showed that the Pt12448 gene was up-regulated in the late induction,and the Pt34354 gene was up-regulated in the early induction.The results of the study will advance the research process of the pathogenic mechanism of P.triticina.
