摘 要:目的:探究铁离子功能化羧甲基甲壳素基微球(Fe@CM-g-AA)作为可吸收止血材料的可行性。方法:结合光学显微镜和电子显微镜观察Fe@CM-g-AA微球的形貌;采用CCK-8和溶血实验评价微球的生物相容性;利用体外酶促降解实验评价微球的生物降解性;利用体外凝血实验评价微球的促凝血性能。结果:Fe@CM-g-AA微球具有完整的球形结构,在生理条件下结构稳定,在体外酶促降解至第9天时,降解超过60%,显示出良好的生物降解性。Fe@CM-g-AA微球对L929和Lewis细胞均无毒性,且溶血率低于5%,具有良好的生物相容性。在凝血实验中,相比于空白对照组,Fe@CM-g-AA微球体外凝血时间从77 s缩短至57 s,能够形成更为牢固的血凝块;活化部分凝血活酶时间(APTT)和凝血酶原时间(PT)均显著缩短(P<0.01)。结论:通过电子束辐照和螯合作用制备了Fe@CM-g-AA微球,该微球能够通过促进红细胞聚集和血液凝血级联反应加速血液凝固,具有作为可吸收止血材料的潜在价值。Objective To explore the feasibility of using the ion-functionalized carboxymethyl chitin-based microsphere(Fe@CM-g-AA)as an absorbable hemostat.Methods The morphology of Fe@CM-g-AA microspheres were observed by op⁃tical microscopy and scanning electron microscopy.The CCK-8 and hemolysis assays were employed to assess the biocom⁃patibility of the microspheres.The in vitro enzymatic degradation experiment was carried out to evaluate the biodegradability of the microspheres.Further,the procoagulant activity of the microspheres was tested by in vitro coagulation experiments.Results Fe@CM-g-AA microspheres exhibited a spherical structure and remained stable under physiological conditions.The microspheres had low hemolysis rates(<5%)and showed no toxicity to L929 and Lewis cells.Moreover,the micro⁃spheres displayed good biodegradability,with the degradation rate over 60%after 9 days.The in vitro coagulation experi⁃ments showed that Fe@CM-g-AA microspheres significantly reduced the clotting time from 77 s to 57 s and formed stron⁃ger blood clots compared with the blank control group.In addition,the microspheres shortened the activated partial throm⁃boplastin time(APTT)and thrombin time(TT)(P<0.01).Conclusion Fe@CM-g-AA microspheres were prepared through electron beam irradiation and chelation.The microspheres can accelerate coagulation by promoting red blood cell aggregation and the cascade reaction of coagulation,demonstrating the potential value as absorbable hemostatic materials.
