广西地区凡纳滨对虾白斑综合征病毒同源重复区基因的比较分析

摘　　要：为了解广西地区对虾白斑综合征病毒(White spot syndrome virus, WSSV)分子变异特征,选取2010-2019年采自广西沿海地区的120份WSSV阳性凡纳滨对虾(Litopenarus vannamei)样品,进行同源重复区ORF75、ORF94和ORF125基因的扩增和测序,并比较分析其重复单元变化差异(Variable number of tandem repeats, VNTR)及单核苷酸多态性(Single nucleotide polymorphisms, SNPs)。结果显示,ORF75的重复单元(Repeat units, RUs)是45bp(RUs-45 bp)和102 bp(RUs-102 bp),RUs数目存在3、4、6、7、8、9类型,最多的是6 RUs型;ORF94的RUs是54bp(RUs-54 bp),RUs数目有0、1、2、3、4、6、8、9、11、12类型,最多的是2 RUs型;ORF125的RUs前两个是72 bp(RUs-72 bp),第3个之后的RUs为69 bp(RUs-69 bp),RUs有3、5、6、8、9类型,最多的是6 RUs型。SNPs分析结果表明,ORF75的RUs-45 bp的第3、15、30、42、44位存在多态性,RUs-102 bp的第83位存在多态性;ORF94的RUs-54 bp的第48位具有多态性,为G、T或C;ORF125的RUs-72 bp不存在多态性,RUs-69 bp(从第1个到倒数第2个)的第2、12、19、22、37、40、48、53、60、63位具有多态性,第2位为T/C,12和22位为G/T,19位为G/A,37、40、48、53和63位为A/G,60位为C/T,出现较多的位点是第12、22、37、60、63位,其中第60、63位多态性最为普遍。结果表明,WSSV广西株同源重复区存在较大变异,序列的RUs数目和SNP存在多样性。To understand the molecular variation of white spot syndrome virus(WSSV)from Penaeus vannamei(P.vannamei)farming in Guangxi,China,we collected 120 WSSV‐positive Penaeus vannamei(P.vannamei)samples during 2010 to 2019 in this region to amplify,clone and sequence the ORF75,ORF94 and ORF125 genes.In addition,the VNTR(Variable number of tandem repeats)and SNPs(Single Nucleotide Polymorphisms)in the highly variable region presented in ORF75,ORF94 and ORF125 were compared and analyzed.The results showed that the RU(repeat units)in ORF75 were 45 bp and 102 bp in length,and the numbers of RU were 3,4,6,7,8 and 9,most of which were 6RU.The RU in ORF94 was 54 bp in length,the numbers of RU were 0,1,2,3,4,6,8,9,11 and 12,most of which were 2RU.While the RU in ORF125 were 72 bp in length in the first two RUs and 69 bp in length from the third RU,the numbers of RUs were 3,5,6,8 and 9,most of which were 6RU.SNPs analysis results demonstrated that the SNPs in ORF 75 were found at positions 3,15,30,42 and 44 of 45 bp RU type,and the RU‐102 bp have SNPs at positions 83.The SNPs in ORF94 was T/G/C at position 48 of RU‐54 bp.The SNPs in ORF 125 are not presented in RUs‐72 bp,but are found at position 2,12,19,22,37,40,48,53,60 and 63 in RU‐69 bp,and the SNPs were T/C at position 2,G/T at position 12 and 22,G/A at position 19,A/G at position 37,40,48,53 and 63,C/T at position 60.More SNPs appeared at position 12,22,37,60 and 63,of which 60 and 63 were most common position.The results suggested that variation region of WSSV strains in Guangxi exert great variation,and there was diversity in the RUs and SNP.
