摘 要:目的建立基于二代测序(next-generation sequencing,NGS)技术对人类白细胞抗原(human leucocyte antigen,HLA)基因进行分型的自动化操作平台,并评估其在HLA常规检测中的应用。方法根据试剂盒操作说明编写相应的计算机程序,利用Hamilton Microlab STARlet全自动液体处理工作站联合96通道小型液体工作站、荧光化学发光分析仪等仪器,实现NGS技术对HLA基因分型过程中文库制备步骤的自动化操作。结果手工法和自动化平台获取的文库片段两者读长基本一致,浓度及质量均符合要求,测序结果一致;而自动化平台操作时间明显少于手工操作。结论建立的NGS自动化操作平台检测HLA基因分型方法可行,值得推广使用。Objective To establish an automated platform for human leucocyte antigen(HLA)genotyping using next-generation sequencing(NGS)technology,and evaluate its application in routine HLA genotyping.Methods The computer programs were written according to the kit operation instructions,and Hamilton Microlab STARlet,96 channel liquid workstation,Fluoroskan Ascent and other instruments were used to accomplish the automatic operation of library preparation in the process of HLA genotyping by NGS technology.Results The reads length and sequencing results of library obtained by automated platforms were consistent with the library obtained by manual,and the concentration and quality of library were acceptable.The operation time of automatic platform was obviously less than manual operation.Conclusion The automated platform for HLA genotyping using NGS technology is feasible and worth popularizing.
