四君子汤对脾虚结肠癌相关巨噬细胞极化的影响研究

摘　　要：目的探讨四君子汤对脾虚结肠癌相关巨噬细胞(TAMs)极化的影响。方法使用利血平建立脾虚小鼠模型,然后小鼠结直肠癌细胞建立小鼠肿瘤模型,待肿瘤长至50mm3左右时,给予四君子汤治疗,观察脾虚小鼠模型是否建立成功、肿瘤抑制率、TAMs分型变化。结果脾虚结肠癌组(Ⅲ)和脾虚结肠癌+中药组(Ⅳ)组脾虚评分均明显高于正常对照组(Ⅰ)和结肠癌模型组(Ⅱ)(P<0.05);而Ⅲ和Ⅳ组,Ⅰ和Ⅱ组脾虚评分无明显差异(P>0.05)。经过四君子汤治疗显示,Ⅳ组肿瘤与结肠癌组和脾虚结肠癌组相比均明显被抑制(P<0.05),同时Ⅳ组脾脏指数明显降低,而胸腺指数较高(P<0.05)。Ⅳ组肿瘤与结肠癌组和脾虚结肠癌组相比M2型巨噬细胞均明显减少(P<0.05)。结论四君子汤对可促进脾虚结肠癌TAMs向M1型转化,进而抑制肿瘤增殖。Objective To explore effect of Decoction of Four Noble Drugs on polarization of colon cancer associated macrophages(TAMs)with spleen deficiency.Methods The paper established mouse model of spleen deficiency with reserpine,and mouse tumor model with mouse colorectal cancer cells.As the tumor grew to about 50mm3,samples were treated with Decoction of Four Noble Drugs.Success of spleen deficiency mouse model,tumor inhibition rate,and changes in TAMs typing were observed.Results Spleen deficiency scores of spleen deficiency colon cancer group(Ⅲ)and spleen deficiency colon cancer+traditional Chinese medicine group(Ⅳ)were significantly higher than normal control group(I)and colon cancer model group(Ⅱ)(P<0.05).However,there was no significant difference in spleen deficiency scores between groupsⅢandⅣ,and groups I andⅡ(P>0.05).After treatment with Decoction of Four Noble Drugs,it was found tumors in GroupⅣwere more significantly inhibited than colon cancer group and spleen deficiency colon cancer group(P<0.05).At the same time,spleen index in GroupⅣwas significantly reduced,while thymus index was higher(P<0.05).Comprison showed between colon cancer group and spleen deficiency colon cancer group,M2 macrophages in GroupⅣwere significantly reduced(P<0.05).Conclusion Decoction of Four Noble Drugs can promote transformation of TAMs from spleen deficiency colon cancer to M1 type,thereby inhibite tumor proliferation.
