摘 要:目的探讨α-MSH调控Bcl-2线粒体途径在桃红四物汤防治激素性股骨头坏死中的作用机制。方法SD大鼠臀肌注射甲泼尼龙琥珀酸钠构建激素诱导相关的股骨头坏死大鼠模型;灌胃给予桃红四物汤干预;并以臀肌注射生理盐水的大鼠为对照。给药8周后取材,体外诱导培养成骨细胞,观察CCK-8,DAPI染色,Western blotting免疫蛋白印迹方法分析α-MSH调控Bcl-2的蛋白表达水平。结果CCK-8检测α-MSH对原代成骨细胞的细胞毒性较低,CCK-8检测α-MSH对激素诱导的原代成骨细胞凋亡显示较强的抗凋亡作用(P<0.05);激素诱导后的原代成骨细胞活力受到抑制,但是α-MSH对激素诱导的原代成骨细胞未显示出明显的促进细胞活力的作用。WB检测相关凋亡指标未显示出明显的α-MSH对激素诱导的原代成骨细胞凋亡的抗凋亡作用。DAPI观察激素处理后可见部分细胞核变小,说明激素可能诱导成骨细胞的凋亡,但是α-MSH未显示出明显的抗凋亡作用。结论α-MSH调控Bcl-2线粒体途径在桃红四物汤防治激素性股骨头坏死中的作用机制,为临床诊疗股骨头坏死提供新的研究思路,并值得进一步探讨其分子机制。Objective To investigate the mechanism of a-MSH regulating Bcl-2 mitochondrial pathway in Taohong Siwu Decoction for prevention and treatment of hormone-induced femoral head necrosis.Methods SD rat gluteal muscle was injected with methylprednone sodium succinate to establish the model of hormoneinduced femur head necrosis.Taohong Siwu decoction was administered by irrigation.Rats injected with normal saline in gluteal muscle were taken as control.After 8 weeks of administration,osteoblasts were induced and cultured in vitro.CCK-8,DAPI staining,and Western blotting method were used to analyze the expression level of Bcl-2protein regulated by a-MSH.Results The cytotoxicity of a-MSH on primary osteoblasts was low as detected by CCK-8,and a-MSH showed strong anti-apoptotic effect on hormoneinduced apoptosis of primary osteoblasts by CCK-8(P<0.05).The apoptosis of primary osteoblasts induced by hormone was inhibited,but a-MSH did not significantly promote the apoptosis of primary osteoblasts induced by hormone.WB showed no significant anti-apoptotic effect of a-MSH on steroid-induced primary osteoblast apoptosis.DAPI showed that some cell nuclei became smaller after hormone treatment,indicating that hormone may induce apoptosis of osteoblasts,but a-MSH showed no obvious anti-apoptotic effect.Conclusion The mechanism of Bcl-2 mitochondrial pathway regulated by a-MSH in the prevention and treatment of hormone-induced femoral head necrosis by Taohong Siwu Decoction provides a new research idea for clinical diagnosis and treatment of femoral head necrosis,and is worth further exploring its molecular mechanism.
