摘 要:糖基化修饰是一种重要的蛋白翻译后修饰,细胞膜上的蛋白一般多为糖蛋白,细胞内也不断发生着不同类型的糖基化修饰反应。糖蛋白参与到宿主的众多生命过程中,包括细胞生长、细胞间相互识别、免疫应答、病毒入侵及炎症反应的发生等。随着蛋白质组学和糖组学技术的不断完善,蛋白的糖基化修饰研究得到越来越广泛的重视。随着生物技术的不断发展,研究方法不断完善,我们对糖基化修饰所带来的蛋白功能与构象的改变也有了越来越清楚的了解。关于糖型分析及糖蛋白上不同糖型比例的问题也随着糖蛋白的检测、分离纯化、糖基化氨基酸位点测定、糖链结构的分析等实验技术的不断完善得到解决。糖基化对多种疫苗候选抗原的免疫原性影响已有许多相关研究,例如乙型肝炎病毒、人类免疫缺陷病毒(human immunodeficiency virus,HIV)、流感病毒以及新型冠状病毒等疫苗抗原/候选抗原都有不同程度糖基化修饰,糖基化修饰对这些疫苗抗原的影响同样非常重要。本文就糖基化研究方法及糖基化对多种疫苗免疫原性的影响进行综述。Glycosylation modification is an important post-translational modification of proteins.Most proteins on cell membrane are glycoproteins,and different types of glycosylation modification reactions constantly occur inside the cell.Glycoproteins are involved in many life processes of the host,including cell growth,mutual recognition between cells,immune response,virus invasion and inflammatory response.With the technology improvement of proteomics and glycoomics,studies on glycosylation and modification of proteins have been paid more and more attention.With continuous development of biotechnology and research methods,we are having clearer understanding of the changes in protein function and configuration caused by glycosylation modification.The problems of glycotype analysis and the proportion of different glycotypes on glycoproteins are being solved with continuously developed experimental techniques such as the detection,separation and purification of glycoproteins,the determination of glycosylated amino acid sites,and the analysis of glycochain structure.There have been many studies on the influence of glycosylation on the immunogenicity of multiple vaccine/candidate antigens.Vaccine antigens/candidate antigens such as hepatitis B virus,human immunodeficiency virus(HIV),influenza virus and novel coronavirus(COVID-19)are all modified with varying degrees of glycoylation.Glycosylation modifications are also important in influencing these vaccine antigens.This article reviews the research methods of glycosylation and the effects of glycosylation on the immunogenicity of various vaccines.
