摘 要:聚β内酰胺主链类似于多肽骨架结构,具有良好的生物相容性,在细胞黏附和细胞选择性生物材料领域具有应用前景。丙烯酰胺(AM)的质子转移聚合是制备聚β内酰胺的简易方法,但是其产物相对分子质量不高、不具力学性能。为此,文中推出先通过AM与1,4-丁二醇二丙烯酸酯(BDDA)的质子转移共聚合制备含双键的β内酰胺预聚物,然后再对其光固化交联来制备具有良好生物相容性和力学性能的可降解β内酰胺交联共聚物。通过核磁共振氢谱和傅里叶变换红外光谱仪对聚合物结构进行表征,通过拉伸性能测试对聚合物力学性能进行研究,通过四甲基偶氮唑盐比色法(MTT法)对细胞在材料上的存活率进行测试。结果表明,聚合物的拉伸强度高达56.6 MPa,断裂伸长率高达56.6%,明显高于高密度聚乙烯、聚丙烯等常用聚合物材料。不仅如此,所制备β内酰胺交联共聚物可极大地促进细胞增长,细胞存活率高达170%。Poly(β-lactam)with polypeptide-like skeleton structure has a good biocompatibility.It has been widely applied as cell adhesion and cell selective biomaterials.Generally,poly(β-lactam)can be synthesized by hydrogen transfer polymerization(HTP)of acrylamide(AM).However,the low molecular weight of the resulted polymer make it show poor mechanical properties.Herein,poly(b-lactam)prepolymers with large amounts of double bonds were firstly prepared by the hydrogen transfer copolymerization of AM and 1,4-butanediol diacrylate(BDDA).Then,the products were further crosslinked by UV,resulting crosslinked b-lactam copolymers.The structures of the resulted polymers before and after crosslinking were confirmed by nuclear magnetic resonance(1H-NMR)and Fourier infrared spectrometer(FT-IR).The mechanical properties and compatibility of the products were also studied by tensile test and methyl thiazolyl tetrazolium(MTT),respectively.The results show that the tensile strength of the polymer is up to 56.6 MPa accompanying with an elongation at break of 56.6%,which is significantly higher than those of commonly used polymers such as high-density polyethylene(HDPE)and polypropylene(PP).Moreover,the cell survival rate for these materials could get up to 170%,showing a good growth-promotion of cell for these materials.
