摘  要:目的探讨Kidd血型溶质载体家族14基因(solute carrier family 14,SLC14A1)在肾透明细胞(kidney renal clear cell carcinoma,KIRC)中的表达及临床意义。方法运用生物信息学分析方法,联合SANGERBOX、GEPIA、UALCAN、STRING等数据库分析和评估SLC 14A1在KIRC中的表达和临床预后价值。结果Kidd血型SLC 14A1基因在4种肿瘤中观察到了显著上调(P<0.05),在22种肿瘤中观察到了显著下调(P<0.05),其中SLC 14A 1在KIRC组织中mRNA表达水平显著低于正常组织(1.35±1.69 vs 3.90±2.47,P=4.7e-36)。生存分析表明,SLC 14A1表达水平与KIRC总体生存率呈显著正相关(P<0.05)。基于单因素Cox和多因素Cox回归结果分析低表达SLC 14A1是KIRC预后危险因素。KEGG富集分析代谢途径通路占主导地位,GO富集分析吞噬体酸化、三价铁运输、转铁蛋白转运占主导地位。结论Kidd血型SLC 14A1基因在多数KIRC中呈现低表达,与KIRC的发生、发展和预后不良相关,有望作为KIRC诊断、预后判断和治疗靶点。Objective To investigate the expression and clinical significance of Kidd blood group solute carrier family 14(SLC 14A 1)gene in kidney clear cell carcinoma(KIRC).Methods Bioinformatics analysis was used to analyze and evaluate the expression and clinical prognostic value of SLC 14A1 in KIRC in combination with SANGERBOX,GEPIA,UALCAN,STRING and other databases.Results Kidd blood group SLC 14A1 gene was significantly up-regulated in 4 tumors(P<0.05),and significantly down regulated in 22 tumors(P<0.05).The mRNA expression level of SLC 14A1 in KIRC tissues was significantly lower than that in normal tissues(tumor1.35±1.69 vs normal 3.90±2.47,P<0.05).Survival analysis showed that the expression level of SLC 14A1 was significantly positively correlated with the overall survival rate of KIRC(P<0.05).Based on univariate Cox and multivariate Cox regression analysis,low expression of SLC 14A1 was a prognostic risk factor for KIRC.KEGG enrichment analysis dominates in metabolic pathways.GO enrichment analysis dominates in phagosome acidification,trivalent iron transport and transferrin transport.Conclusion The low expression of Kidd blood group SLC 14A1 gene in KIRC is associated with the occurrence,development and poor prognosis of KIRC,which is expected to be used as a possible target for KIRC diagnosis,prognosis judgment and treatment.
