摘 要:本研究以具有高水平禽滑液囊支原体(MS)特异性母源抗体商品蛋鸡作为实验动物,评估了母源抗体对MS减毒活疫苗免疫效果的影响。分别在7日龄、14日龄、21日龄和28日龄免疫MS-H株活疫苗,每周采集腭裂拭子样品,通过荧光定量PCR方法(qPCR)检测MS-H疫苗株基因组拷贝数,以评估疫苗株在体内的定植情况。在28日龄免疫组免疫后5周对各试验组用MS强毒株进行攻毒,连续观察4 w,每周采集腭裂拭子通过qPCR方法检测攻毒菌株基因组拷贝数,以评价疫苗免疫对野毒感染的阻断效果,最后扑杀并剖检所有试验鸡,观察和记录特征性病变。结果表明,MS特异性母源抗体在3周龄时基本降至阴性水平,早期高母源抗体水平对疫苗毒株的定植会产生一定程度的影响,但不同时间免疫组疫苗株均能有效定植,且对强毒株攻毒的保护效果影响不显著。本研究的相关结果对于指导生产中科学制定MS活疫苗的免疫程序具有重要的参考意义。In this study,commercial laying hens with high levels of maternally-derived antibodies(MDAs)againstMycoplasmasynoviae(MS)were used as experimental animals to evaluate the effect of MDAs on the efficacy of an attenuated live vaccine.The live MS vaccine(MS-H strain)was administered to chickens at 7,14,21 and 28 days of age,respectively.Choanal cleft swabs were collected weekly and the genomic copy number of MS-H vaccine strain was examined in real-time quantitative PCR(qPCR)to evaluate invivo colonization.At 5 weeks post thefinal vaccination,all chickens were challenged with a virulent MS strain and observed for 4 weeks.When chickens were terminated,lesions were observed and recorded.The results showed that the MS-specific MDAs decreased to negative level at 3 weeks of age and the early high MDA level had a certain influence on the colonization of the vaccine strain.However,the vaccine strains effectively colonized in the immunization groups vaccinated at different times and the protection against the virulent challenge was not significantly influenced.These results provided important reference for guiding the immunization procedure of MS live vaccine in the production.
