摘 要:铁死亡作为一种新型非程序性细胞死亡方式,已被证实与食管癌的发病密切相关。通过铁死亡途径干预食管癌细胞的增殖,为食管癌的精准治疗提供了新方法,具有较好的临床应用价值。然而,铁死亡途径的调节通路较多,食管癌的发生发展亦具有复杂性,故铁死亡在食管癌中的作用机制尚未完全明确。近年来,铁死亡在食管癌中的研究已取得重大进展,但仍有一些问题亟待解决:在食管癌中,铁死亡与其他细胞死亡形式之间是否存在相互调控作用还有待评价;铁死亡对食管癌的具体干预阶段及靶点仍需探索;现有研究多为理论或实验研究,真正把铁死亡理论用于食管癌临床治疗的还很少;目前,已被证实的一些铁死亡诱导剂有待临床进一步验证。今后可进一步从代谢、免疫和遗传学的角度更全面地揭示和分析铁死亡在食管癌发病中的作用机制,探寻能够反映食管癌细胞对铁死亡敏感的生物标记物和治疗靶点,发现新的铁死亡诱导剂并进行临床评价,提高食管癌诊治水平。Ferroptosis,as a novel non-programmed cell death modality,has been shown to be closely related to the pathogenesis of esophageal cancer.Intervening in the proliferation of esophageal cancer cells through ferroptosis provides a new method for the precision treatment of esophageal cancer and has good clinical application value.However,the mechanism of ferroptosis in esophageal cancer has not been fully understood,due to the fact that there are many regulatory pathways in ferroptosis,and the occurrence and development of esophageal cancer are complex.In recent years,significant progress has been made in the research of ferroptosis in esophageal cancer,but there are still some problems that need to be solved urgently:whether there is a mutual regulatory effect between ferroptosis and other forms of cell death in esophageal cancer remains to be evaluated;The specific intervention stages and targets of ferroptosis for esophageal cancer still need to be explored.Most of the existing studies are theoretical or experimental studies,and there are very few that really apply ferroptosis theory to the clinical treatment of esophageal cancer.Some of the currently proven ferroptosis inducers are pending further clinical validation.In the future,the mechanism of ferroptosis in the pathogenesis of esophageal cancer can be more comprehensively revealed and analyzed from the perspectives of metabolism,immunity and genetics,and biomarkers and therapeutic targets that can reflect the sensitivity of esophageal cancer cells to ferroptosis,and new ferroptosis inducers can be discovered and clinically evaluated,to improve the diagnosis and treatment of esophageal cancer.
