摘 要:目的:通过生物信息学技术分析银屑病皮损与正常组织的基因芯片数据,筛选银屑病的差异表达基因(differentially expressed genes,DEGs),探讨银屑病的病理机制,预测治疗银屑病的潜在中药。方法:从基因表达数据库(gene expression omnibus,GEO)提取数据集GSE161683、GSE193128、GSE137218原始数据,使用R语言Limmar包筛选DEGs。利用DAVID数据库对DEGs进行功能和信号通路的富集分析,通过String数据库构建蛋白互作网络,Cytoscape软件及其插件筛选关键基因。将关键基因与Coremine Medical相互映射,筛选治疗银屑病潜在的靶向中药。结果:筛选出DEGs 474个,其中上调DEGs的通路主要富集在白细胞介素-17(interleukin-17,IL-17)、核苷酸寡聚化结构域样受体(nucleotide oligomerization domain like receptor,NLR)、病毒感染等信号通路。STAT1、RSAD2、IFIT3、IFI44L、IFIT1、CXCL10、MX1、IFI44、OASL、OAS1、OAS2、OAS3为筛选的关键基因。预测出治疗银屑病的潜在中药有黄芩、水牛角、大青叶、当归、赤芍、牡丹皮、胆南星、半夏、何首乌、墨旱莲、鳖甲胶、淫羊藿、鹿茸等。结论:筛选出的12个关键基因可能与银屑病皮损的发生相关,皮损发病主要涉及免疫炎症、角质形成细胞的异常增生、抗病毒感染等机制,筛选出的中药为银屑病治疗的潜在靶向中药。Objective:To analyze the gene chip data of psoriasis lesions and normal tissues using bioinformatics technology and screen them to select differentially expressed genes(DEGs)in psoriasis,explore the pathological mechanism of psoriasis,and predict potential traditional Chinese medicine treatments for psoriasis.Method:Extract the raw data of GSE161683,GSE193128,and GSE137218 from the gene expression database(GEO),and use R language Limmar package to screen for DEGs.Using the DAVID database for functional and signaling pathway enrichment analysis of DEGs,constructing protein interaction networks using the String database,and screening key genes using Cytoscape software and its plugins.Map key genes with Coremine Medical to screen potential targeted traditional Chinese medicine for the treatment of psoriasis.Result:474 differentially expressed genes(DEGs)were identified,and the pathways upregulating DEGs were mainly enriched in interleukin-17(IL-17),nucleotide oligomerization domain like receptors(NLR),viral infections,and other signaling pathways.STAT1,RSAD2,IFIT3,IFI44L,IFIT1,CXCL10,MX1,IFI44,OASL,OAS1,OAS2,OAS3 are the key genes screened.Potential traditional Chinese medicines for the treatment of psoriasis have been predicted to include Huangqin(scutellaria baicalensis),Shuiniujiao(buffalo horn),Daqingye(folium Isatidis),Danggui(angelica sinensis),Chishao(red peony root),Mudanpi(peony bark),Dannanxing(bile arisaema),Banxia(pinellia ternate),Heshouwu(polygonum multiflorum),Mohualian(eclipta),Biejiajiao(turtle shell gelatin),Yinyanghuo(epimedium),Lurong(cartialgenous),etc.Conclusion:The 12 key genes screened may be related to the occurrence of psoriasis skin lesions,which mainly involve mechanisms such as immune inflammation,abnormal proliferation of keratinocytes,and antiviral infection.The screened traditional Chinese medicine is a potential targeting traditional Chinese medicine for the treatment of psoriasis.
