摘 要:0引言小麦黄花叶病是我国冬麦区最重要的病毒病害,其病原是小麦黄花叶病毒(wheat yellow mosaic virus,WYMV)[1],可引起小麦叶片黄化,分蘖减少,株型矮缩,后期麦穗短小,发育不良[2],一般造成小麦减产10%~30%,严重的达70%以上[3]。自20世纪90年代以来,该病害每年发生面积都在66.7万公顷以上,主要分布在我国河南、江苏、山东、安徽、陕西、湖北、四川等省,呈逐年蔓延加重趋势[3-4]。Wheat yellow mosaic virus(WYMV)disease,the most important viral disease in the winter wheat region of China,is more and more serious in recent years.The pathogen,WYMV,is transmitted by Polymyxa graminis.Investigating the connection between wheat cultivar’s resistance to WYMV and the amount of P.graminis in root is instructive,as it is beneficial to the understanding of the mechanism of wheat cultivar’s resis-tance and can offer guidance for the breeding of cultivars.Based on the rDNA of P.graminis,a pair of specific primers were designed.The SYBR Green I real-time PCR reaction system was established and optimized.We evaluated 70 wheat cultivars according to their disease severity in field disease nursery and examined whether P.graminis existed in their root using PCR.Four cultivars,which exhibited uniform disease severity,were chosen from a collection of cultivars with similar disease severity,and the amounts of P.graminis in their roots were determined using real-time PCR.Results showed that among the 70 cultivars,32 exhibited level 0 of disease severity,9 exhibited level 1,7 exhibited level 2 and 22 exhibited level 3.P.graminis was identified from the root of all these 70 cultivars using PCR.Real-time PCR showed that the wheat cultivar that exhibited higher disease severity had a greater number of P.graminis in their root.The disease severity of wheat cultivar presented a positive relationship with the amount of P.graminis in root.The wheat cultivars that exhibited level 3 of disease severity had the greatest number of P.graminis in their root.But,some wheat cultivars that exhibited level 0 of disease severity had a greater number of P.graminis in their root.These results indicated that the resistance of wheat cultivar to WYMV was related to the resistance to P.graminis in their root.
