升阳益胃汤对慢性胃炎小鼠胃组织病理变化 及p38、ERK蛋白表达的影响

摘　　要：目的:观察升阳益胃汤对慢性胃炎小鼠胃组织病理变化及细胞外信号调节蛋白ERK和丝裂原激活蛋白p38表达的影响。方法:健康雄性小鼠40只随机分为正常组、模型组、升阳益胃汤组和西药组,每组10只。以2%水杨酸钠灌胃结合饥饿饱餐因素制备慢性胃炎小鼠模型。成模后给药组分别予升阳益胃汤(10 g/kg)、维酶素悬浊液(0.33 mg/kg)灌胃30 d。取胃标本病理制片后观察胃黏膜及组织病理变化,免疫组化法测定各组小鼠胃组织p38、ERK表达。结果:正常组小鼠胃组织未见炎性病理改变,模型组可见胃黏膜炎症表现,西药组炎症程度减轻,升阳益胃汤组炎症程度明显减轻。免疫荧光结果显示模型组小鼠胃组织p38、ERK表达水平高于正常组(P<0.05),升阳益胃汤组p38、ERK表达水平低于模型组(P<0.05),但高于正常组(P<0.05)。结论:升阳益胃汤可能通过调节p38、ERK表达水平减轻慢性胃炎小鼠的胃黏膜病理损伤。Objective:To explore the effects of Shengyang Yiwei Decoction on pathological changes of gastric mucosa and expressions of p38、ERK protein in chronic gastritis mouse.Methods:40 healthy male mice were divided into normal group,model group,Shengyang Yiwei Decoction group and Western medicine group with each 10 mice.The model of chronic gastritis was established by gavage 2 mL of 2%sodium salicylate every day for 8 weeks,each group was given corresponding medicine by gavage for 30 days,and gastric specimens were taken for HE staining,gastric tissues were observed with light microscope.The expressions of gastric tissues p38 and ERK protein in mice was measured by immunofluorwescence Results:No inflammatory pathological changes were found in the gastric mucosa of normal mice;In the model group,gastric mucosa inflammation was observed;The degree of inflammation in Western medicine group was reduced;The degree of inflammation in Shengyang Yiwei Decoction group was significantly reduced.Immunofluorwescence detection results showed that the expressions of gastric tissues p38 and ERK protein in model group were higher than those of normal group,p38 and ERK protein expressions of Shengyang Yiwei Decoction group were lower than those of model group,but that were higher than those of normal group.Conclusion:Shengyang Yiwei Decoction can reduce the pathological changes of mucosa in chronic gastritis mice by regulating p38 and ERK protein expressions.
