摘  要:为了探究2020年江苏省扬州市江都区某鹅场发生的1起以肝脏和脾脏出血为典型病变特征的急性传染病病因,本试验采用反转录聚合酶链反应(RT-PCR)方法检测病死鹅肝脏和脾脏研磨液中禽呼肠孤病毒(ARV)的感染情况,将聚合酶链反应(PCR)阳性病料进一步进行病毒的分离、鉴定、血凝性和致病性测定及遗传进化分析。结果显示,鸡胚中分离的病毒为ARV,将其命名为ARV-JSJD-2020;该病毒分离株接种鸡胚24 h后即可致其死亡;能感染Vero细胞产生细胞病变;没有血凝活性;鸡胚半数致死量(ELD50)为10^(-4.166)/0.2 mL;动物回归试验中雏鹅24 h即出现死亡现象,肝脏主要病变为局灶性坏死;σC基因序列分析显示,该分离株分类上属于水禽源呼肠孤病毒分支,但与目前疫苗株的亲缘关系较远。结果表明,本试验分离得到的ARV-JSJD-2020可以为后续鹅源ARV的防控和疫苗的研发提供参考依据。In order to investigate the etiology of an acute infectious disease with typical liver and spleen hemorrhagic lesions that occurred in a goose farm in Jiangdu District,Yangzhou City,Jiangsu Province in 2020,this study used reverse transcription-polymerase chain reaction(RT-PCR)to detect the infection of avian reovirus(ARV)in liver and spleen homogenates of diseased geese.Positive polymerase chain reaction(PCR)samples were further subjected to virus isolation,identification,hemagglutination,pathogenicity testing,and genetic evolution analysis.The results showed that the isolated virus from chicken embryos was identified as ARV and named ARV-JSJD-2020.The isolated strain caused death in chicken embryos 24 hours post-inoculation,induced cytopathic effects in Vero cells,exhibited no hemagglutination activity,and had a median embryo lethal dose(ELD 50)of 10^(-4.166)/0.2 mL.In animal regression tests,goslings exhibited mortality within 24 hours,and the main pathological change in the liver was focal necrosis.Sequence analysis of theσC gene indicated that the isolated strain belonged to the branch of ARV from waterfowl sources,but it showed distant genetic relationship with current vaccine strains.These results suggest that ARV-JSJD-2020 isolated in this study can provide a reference for the prevention and control of goose-origin ARV and the development of vaccines.
