益气化瘀中药通过干预Sema4D及其受体对糖尿病大鼠创面血管新生的影响

摘　　要：目的探讨益气化瘀中药通过干预Sema4D及其受体对糖尿病大鼠创面血管新生的影响。方法将150只大鼠随机分为正常组、糖尿病模型组、益气组、化瘀组、益气化瘀组。使用链脲佐菌素腹腔注射和外科方法复制糖尿病大鼠皮肤缺损模型,运用免疫组化图像分析技术,分别检测各组第3、7、14天创面Sema4D以及其受体PLexinB1和VEGF的表达水平,并观察创面新生血管情况。结果糖尿病溃疡模型组创面Sema4D、PLexinB1、VEGF的表达水平均明显低于正常组(P<0.001);益气化瘀组创面Sema4D、PLexinB1、VEGF的表达水平高于糖尿病溃疡模型组(P<0.001),明显高于化瘀组(P<0.001),且在第14天明显高于益气组(P<0.001);在第3、7天,糖尿病溃疡模型组创面新生微血管密度(MVD)明显低于正常组(P<0.001),益气化瘀组和益气组创面MVD明显高于糖尿病溃疡模型组(P<0.001),在第14天,糖尿病溃疡模型组创面MVD明显高于正常组(P<0.001),各治疗组均明显低于糖尿病溃疡模型组(P<0.01)。结论益气化瘀中药能通过提高创面Sema4D及其受体的表达水平,增加VEGF的表达,促进血管新生,从而加速创面愈合。Objective To investigate the effect of Chinese herbs of replenishing Qi and resolving Stasis on wound angiogenesis in diabetic rats by interfering with Sema4D and its receptor.Methods 150 rats were randomly divided into five groups:normal group,diabetic model group,Replenishing Qi treatment group,Resoving Stasis treatment group,and Yiqi Huayu Decoction group(replenishing Qi and resolving Stasis treatment group).Diabetes model was established by peritoneal injection of streptozotocin and skin ulcers were made by surgery method in rats.Immunohistochemical image analysis was used to detect the expression levels of Sema4D,PLexinB1 and VEGF at the 3rd,7th and 14th day in each group respectively,and observe the situation of wound neovascularization.Results The expression levels of Sema4D,PLexinB1 and VEGF in diabetic ulcer model group were significantly lower than those in normal group(P<0.001).The expression levels of Sema4D,PLexinB1 and VEGF of Yiqi Huayu Decoction group were higher than those of diabetic ulcer model group(P<0.001),significantly higher than those of resolving Stasis treatment group(P<0.001),and significantly higher than those of replenishing Qi treatment group on day 14(P<0.001).On the 3rd and 7th day,the new microvascular density(MVD)of the diabetic ulcer model group was significantly lower than that of the normal group(P<0.001),the wound MVD of the supplementing Qi and removing blood stasis groups was significantly higher than that of the diabetic ulcer model group(P<0.001),and on the 14th day,the wound MVD of the diabetic ulcer model group was significantly higher than that of the normal group(P<0.001).All treatment groups were significantly lower than diabetic ulcer model group(P<0.01).Conclusion Chinese herbs of replenishing Qi and resolving Stasis can enhance the expression level of Sema4D and its receptor,increase the expression of VEGF,promote angiogenesis and accelerate wound healing.
