摘  要:目的探讨地黄断面颜色与化学成分含有量及药理活性之间的关系。方法收集30批地黄样品,按断面颜色进行分类。采用HPLC测定梓醇和地黄苷D的含有量;采用电子眼技术提取断面颜色的L*,a*,b*值进行量化测定;采用相关性分析法分析成分含有量与颜色量化值之间的相关性,并对不同断面颜色地黄的抗氧化和降血糖活性进行体外评价。结果随着地黄断面颜色的逐渐加深,梓醇和地黄苷D含量逐渐降低,其中断面黑色梓醇含量分布范围在0.1%~0.85%,地黄苷D含量分布范围为0.07%~0.16%,而断面棕黄色梓醇含量分布在1.03%~2.01%,地黄苷D含量分布在0.12%~0.2%之间。电子眼结果显示,梓醇含有量与断面的色度值L*显著的正相关(P<0.05),与b*呈极显著正相关(P<0.01);地黄苷D含有量与色度值L*呈显著的正相关(P<0.05)。活性测定结果表明,随着断面颜色的加深,地黄抗氧化及降血糖作用呈现一定的波动,总体上,断面黑色的地黄抗氧化和降血糖作用较强。结论地黄断面颜色与地黄成分和药理活性具有一定的相关性。Objective To investigate correlation of the section color,chemical constituents content and pharmacological activities of Radix Rehmanniae.Methods A total of 30 batches of Radix Rehmanniae were collected and classified by section color.The contents of catalpol and rehmannioside D were measured by high performance liquid chromatography(HPLC);The L*,a*and b*values of section color were measured taking electronic-eye technique,and the correlation between chemical constituents content and section color chromaticity value were analyzed,meantime,the antioxidant and hypoglycemic activities in vitro of Radix Rehmanniae of different section color were evaluate.Results Along with the section color gradually being deepened,the contents of catalpol and rehmannioside D of Radix Rehmanni were lower,in which,the contents of catalpol and rehmannioside D ranged from 0.1%~0.85% and 0.07%~0.16% in Radix Rehmanniae of section color black respectively.However,there ranged from 1.03%~2.01% and 0.12%~0.2% in section color brown yellow separately.Results of electronic-eye indicated that the content of catalpol was significant positively correlated with the L*value of section color(P<0.05),and extremely significant positively correlated with the b*value(P<0.01).Similarly,the content of rehmannioside D was significant positively correlated with the L*value(P<0.05).Furthermore,the results of activity evaluation displayed the antioxidant and hypoglycemic activities.
