中医滋阴潜阳法对多囊卵巢综合征伴胰岛素抵抗大鼠PI3K/Akt信号通路的影响

摘　　要：目的探讨中医滋阴潜阳法对多囊卵巢综合征伴胰岛素抵抗大鼠的PI3K/Akt信号通路的影响。方法以来曲唑加高脂饲料诱导建立多囊卵巢及胰岛素抵抗双重大鼠模型。采用中医滋阴潜阳法配合达英-35及二甲双胍干预治疗,用免疫荧光法测定卵巢PI3K、AKT通路信号,以实时荧光定量PCR检测卵巢PI3K、Akt途径信号分子mRNA表达及PI3K、Akt、IRS-1途径信号分子蛋白表达。结果中医滋阴潜阳方药能显著增加卵巢PI3K、AKTmRNA相对表达量(P<0.05),并能显著增加卵巢PI3K、AKT蛋白表达。结论中医滋阴潜阳法可提高PI3K/AKT通路信号表达水平,改善多囊卵巢综合征的高雄激素血症及高胰岛素血症。Objective To analyze the effect of traditional Chinese medicine Nourishing Yin and Astringing Yang method on the PI3K/Akt signaling pathway in rats with polycystic ovary syndrome and insulin resistance.Methods Establishment of a rat model of polycystic ovary and insulin resistance induced by the addition of high fat diet and trazole.Adopting the traditional Chinese medicine method of nourishing Yin and suppressing Yang,combined with the intervention treatment of Diane-35 and metformin,Immunofluorescence assay was used to measure ovarian PI3K and AKT pathway signals,and Real time fluorescence quantitative PCR detection of ovarian PI3K and Akt pathway signaling molecule mRNA expression and PI3K and Akt pathway signaling molecule protein expression.Results The traditional Chinese medicine formula for nourishing Yin and suppressing Yang can significantly increase the relative expression levels of PI3K and AKT mRNA in the ovaries(P<0.01);And significantly increased the expression of ovarian PI3K and AKT proteins.Key word:nourishing Yin to suppress Yang,Yin is not most replete and Yang emerges prematurely,Polycystic ovary syndrome with insulin resistance,PI3K/Akt signaling pathway.Conclusion The method of nourishing Yin and suppressing Yang in traditional Chinese medicine can increase the expression level of PI3K/AKT pathway signal,improve the hyperandrogenism and hyperinsulinemia in polycystic ovary syndrome.
